Rice is the largest crop in our country. Its output and quality are influenced by the quality of the seed and the purity of the seed is the most troublesome problem in the control of seed quality. SDS-PAGE and IEF techniques were applied to compare and analyze five hybrid rice and their three lines and nine different F1 hybrids, which are widely cultivated in Fujian province at present, A simple, rapid and economical method was selected for rice purity identification and a standard procedure was established. At the same time, the purity of several hybrids was also analyzed, discussing its practicability and prospect. Major results were as follows:1. SDS-PAGE technique was applied to analyze alcohol-soluble proteins and embryo proteins on five different hybrid rice and their three lines. Experiments showed that the polymorphism of alcohol-soluble proteins was low and there were no differences in the same hybrid rice and their three lines. So it could not be used for the authenticity and purity assessment. But rich polymorphism and significant differences were found in embryo proteins. Therefore, it could be used to differentiate their parents and other seeds from hybrid F1. Though we sampled in weight not individual, it could also be considered as the differences of varieties. So the embryo proteins could be used for purity identification in theory.2. IEF technique was applied to analyze the alcohol-soluble and embryo proteins on different hybrid rice and their three lines. Experiments showed that the bands of alcohol-soluble and embryo proteins among different hybrid rice and their three lines varied much, so it could be used to identify different samples of the same group and different groups.3. The salt-soluble of four different hybrid rice and their three lines were analyzed based on SDS-PAGE. The results showed that the polymorphism of salt-soluble proteins was high.
