Establishment Of Cell Suspension Cultures From Different Wheat Cultivars With Resistance To Leaf Rust

In this experiment,wheat cultivar lovrin10, 5389 and Lr26 were cultured to study the effect of different mediums on callus induction and the gain of embryonal callus and establish the cell suspension culture line of wheat cultivar lovrin10.The plant regeneration from the cell suspension culture line was also studied. The main results are as follows:1.Effect of different plant growth regulators on callus induction were studied. MS+2,4-D2.00~3.00mg/L culture medium can induce wheat cultivar lovrin10, 5389 to produce the highest ratio of callus, while Lr26 in MS+2,4-D4.00mg/L can gain the same result.2.Effect of different plant growth regulators on callus subculture were studied.The concentration and the proportion of plant growth regulators in subculture medium was regulated with the changing of the callus state. For the eugonic callus we can be reduced the concentration of 2,4-D and increased the concentration of KT properly,while the slow-growing callus need to increase the concentration of 2,4-D and reduce the concentration of KT properly, but the dosage of plant growth regulators can not be too low.By doing this, the embryonal callus of wheat cultivar lovrin10 has been obtained and the embryonal callus of wheat cultivar 5389 could be induced in the future.3.The effect of various concentrations of several plant growth regulators on the cell suspension culture line was studied.and the better concentrations were confirmed.4.The product of the cell suspension culture line was not affected by Casein hydrolysates acid, Glutamine, Asparagine and Malt extract ,but the state of the suspension cell depended on them.5.The stable cell suspension culture line of wheat cultivar lovrinlO was established by choosing the better culture conditions.The growth curves of suspension cell and the cell viability were assayed,and the results confirmed that the interval of suspension cell cultures is 7 days. The plant regeneration was abtained from it.In a word,The cell suspension culture line of wheat luofulin10 was gained and it provided a good system for further studying how the elicitor can induce the defense reaction of host plant.
