Rice long glume (lg) mutant resulted from a natural mutation of flower organ, the mutant have two long glumes which is longer than the fruit total lenghth, and the other parts of flower organ are norm. Genetic analysis indicates that the mutant phenotype is controlled by a single recessive gene. The flower organ development model of dicotyledonous already have the moulding. The monocotyledon ,typical is rice, the model of flower organ development still has not the verdiction, glume is corresponding to the calyces or petal of the monocotyledon ?or corresponding to others? So, there are significance to study the rice mutant long glume in order to perfect and reinforce the flower organ development model of monocotyledon. This article major in the map-based cloning method to find the long glume gene. There are genetics separating colony of the mutant for mapping, one is g/japonica colony 2000 lines, the other is g/ZF802 colony 1300 lines, by the linkage genetics analysing, we use the molecule marker, for example STS marker, CAPS marker, SSR marker, having limited the gene between two close molecule markers on this chromosome. The following results were obtained: 1.During the course of mapping, we designed the efficiency molecule marker add up to 98 couples, thereinto, 41 couples of STS, 35 couples of CAPS, 19 couples of SSR, 3 couples of DCAPS. 2.Long glume gene had been located on the short arm of choromsome 7. 3.At the end,the lg gene had been located on the 120 region of one BAC. 4.Gene predicting during the 120 kb district show that exists 35 candidate genes, selected 5 transcript factors,2 kinases and 1 F-box gene for sequencing by using its PCR products or ligated medium vector pGEM-T-easy. The results show the change of amino acid resulting from single nucleotide acid differentia. 5.Some districts exist recombination hotspot.
