Abstract Effect of exogenous hormones and other factors of frequency induction of wheat (Triticum aestivum L.) somatic embryos is studied. The calli were initiated from different kinds.The following results are acquired:1 .Concentration of 2,4-D in media is necessary for induction of embryogenic calli with the different frequency between the different genotype. The most appropriate concentration of 2,4-D is at 2.0~4.0mg/L.The embryogenic calli incubated in the dark produced more embryogenic calli than incubated in the light conditions.2.Jinnong211 incubated in the MS basal medium supplemented with 2.0mg/L 2,4-D plus 0.5-1.0mg/L KT and decrease the concentration of 2,4-D to 1.0mg/L in the forty-fifth day stimulates the somatic embryogenesis as well as Nongda981 incubated in the MS basal medium supplemented with 2.0mg/L 2,4-D plus 0.5mg/L KT and decrease the concentration of 2,4-D to 1 .0mg/L in the fiftieth day; and the percentage of somatic embryogenesis induction reach to at least 70%.3.It is high on somatic embryogenesis derived from culture in MS basal medium supplemented with 30g/L mannitol, as shown by Jinnong 211 or Nongda 981 were cultured on MS basal medium supplemented with 300mg/L Glu or 500mg/L plus 300mg/L CH respectively. Subculture frequently hold on more high somatic embryogenesis. at intervals of 5-10 days once subculture prevent from browned.4.During the maturation of the somatic embryo in addition of 0.2~0.5mg/L ABA produce somatic embryo simultaneous and improve the number of embryoid on every calli.5.The distinction of embryogenic calli and non- embryogenic calli is obvious observed through microscope: the nucleus of embryogenic calli is bigger; tincted cell'colour is more deeper ;and more cell morphologic is different from non-embryogenic call.
