The bulb of Amaryllis vittata was used explant to establish the technique system of tissue culture. With orthogonal design and analysis of variance, we wanted to find out the best medium composition and the culture process in Amaryllis vittata, and to find out effective measures to prevent the browning of bulb explants. The results were as folllows: 1) Temperature for culture:25±1℃.16 hours illumination every day ,and about 3000 Lux illumination intensity. The pH of medium was adjusted to 5.6-5.8. 2) As inoculation explant, leaves of basal bulb in Amaryllis vittata was better than the other parts of the bulb. 3) The best callus initiation medium was MS+BA 2.0mg/L+NAA1.0mg/L+AC1.0 mg/L. After 30 days culture, the rate of initiation could reach 86.4%. 4) The induction of regenerate shoots : MS+BA2.0mg/L+NAA0.1mg/L+AC 1.0g/L was the best medium. After 65 days culture, the multiplication rate of regenerate shoots could reach 6.26. 5) Rooting culture:For rooting culture,MS+BA0.5mg/L+NAA1.0mg/L was the best medium with rooting rate 100% and average length of root 2.86cm. 6) Active carbon was effective for preventing the explants from browning to a certain extent,1.0g/L was the best initiation concentration. 7) Transplantion: For nursling of plantlets, substrate with the soil of the rotten leaves and sand by 3 to 1 was the optimal. The survial rate of plantlets could be up to 100%, after transplanting soil.
