In this research, the Chinese yam of the DaChi Longyan varieties named CunJinShu as the test material, by using tissue culture techniques, the yam induction of multi buds rooting rapid, propagation,tissue culture seedling transplanting, browning inhibition of tissue culture seedlings and the establishment of sterile propagation system etc were studied, and the key technologies of establishing plant regeneration breeding system on yam were discussed, which provided technical support for yam tissue culture breeding propagation. The results showed as follows:1.Used the Cun JinShu tender stems (with internodes)as explants, with MS as the basic medium, sugar concentration of 3%, the most appropriate hormone combination yam multiple shoot induction was 6-BA0.5mg/L+NAA0.1-0.5mg/L, the time of yam induction of multi buds was short, with a larger number of buds formation and the buds were strong activity.2.During the rooting rapid propagation and transplanting test of tissue culture seedlings, the best medium culture for rooting rapid propagation was 6-BA0.2mg/L +NAA0.2-0.5mg/L, the stem of plants was healthy,the color of leaf was green. The transplanting substrates of tissue culture seedling was perlite and humus soil with mixture 1:1, and with 5 days training time, could improved the transplanting survival rate of seedlings.3.In the yam's tissue culture, the browning of linyuzi explant was the most serious,so the stem with internode as a suitable explants. In the process of induced synthesis cultivation bud differentiation, pretreated with 100mg/L of VC soaking 30min, helped to improve its inhibitory effect on the browning reaction, with 150mg/L of PVP solution could reduce browning of plants, to improve the survival rate of multi buds.The yam tissue culture's rapid propagation could be used in MS medium without agar. In the rapid propagation period, it was diffcult for the quinone materials to make an effective role in the liquid solution, which not only can improve the growth potential and survival rate in tissue culture, propagation coefficient of expansion, but also can save production costs during the process of yam tissue culture seedlings without sugar.4.Added to the culture medium with 25mg/L sodium benzoate solution can effectively reduce the contamination rate of explants, which not affecting the explants induction of differentiation rate, could improve the growth environment of explant,and help improve some of the established role in promoting the establishment of sterile propagation system.
