The materials of this thesis are leaves and stems with Carya illinoensis seedlings and adult trees.The article comprehensively and systemically studied on explant choosing, optimum culture medium, plant growth regulators and its ratio, leaves culturing, leaves culturing, browning of explant, Callus induction and so on. Primary results go as follows:1. It is supposed to be that the material of Carya illinoensis explanting cultivating is juvenile leaves and DKW medium can make higher callus induction rate. Using75%alcohol disinfecttion10s rapidly and soak0.1%HgCl24min can reduce the contamination rate. Each bottle of medium inoculation one or two explants also can reduce the contamination rate.2. In Carya illinoensis leaves tissue culture, using DKW+NAA0.15mg/L+2,4-D0.5mg/L+6-BA1.5mg/L can decrease the browning rate. And use DKW+NAA0.1mg/L+2,4-D1.5mg/L+6-BA2.0mg/L can improve callus induction rate. We need add other ingredients as follows:sucrose30g/L, agar lOg/L and adjust pH=5.8.3. The routine disinfection explant inoculated the medium only with sucrose and agar two days, then transferred it into DKW+NAA0.10mg/L+2,4-D1.5mg/L+6-BA2.0mg/L can effectively reduce the rate of browning in Carya illinoensis leaves tissue culture.4. Added1.0g/L AC,1.5mg/L Na2S2O3or0.02mg/L PVP in medium can reduce the rate of browning of the explants, and don't affect callus induction. Added Na2S2O3make the lowest browning rate, PVP make the hightest callus induction. At the same time, adding GA3in the medium can inhibit the callus induction5. Using400mg/L PVP or200mg/L Na2S2O3soak the materials or before inoculation can effectively reduce the browning rates. And make the medium completely shading after inoculation also can reduce the browning rate.6. The best hormone combinations and ratio for Carya illinoensis proliferation of callus culture is6-BA0.5mg/L+NAA0.1mg/L7. In Carya illinoensis stem tissue culture experiments, we selecting juvenile stem as materials, and use WPM+NAA1.0mg/L+6-BA3.0mg/L as basic medium, at the same time add30g/L sucrose and lOg/L agar in it adjust pH=5.8.8. For reducing the browning rates in Carya illinoensis stem tissue culture, the method as follows:add1.0mg/L AC or90mg/LPVP or7mg/L Na2S2O3in the medium; using400mg/L PVP or300mg/L Na2S2O3soak the materials.
