Xinjiang is the base of breeding of fine wool sheep. Many factors have brought the decrease ofbreeding of fine wool sheep in Xingjiang in resent years. The input of wool is very high, and the woolproduction has developed towards light,softness ,high quality facets all of the world. The quality of woolhas also been reformed deeply in world, which is mainly showed by the fine of wool. Considering theabove-mentioned condition, the fine wool sheep should be bread to adapt to the requirement of the market.This experiment was divided into two parts. Firstly , the genetic polymorphism of 9 microsatellites inHigh Quality Merino Sheep were studied by means of PCR and polyacrylamide gel (8%)to reveal thegenetic diversity of High Quality Merino Sheep.Number of alleles ,average effective number ofalleles(Ne) of sheep breeds were counted. Allele frequencies of nine microsatellites,polymorphisminformation content(PIC) and mean heterozygosity(h) were calculated for sheep breeds by UVI-08software.The results showed that 8 of 9 microsatellites loci were high polymorphic, but G54279 weremedium polymorphic. So the 8 of microsatellites loci were effective markers for analysis geneticpolymorphism among High Quality Merino Sheep.The average PIC(0.720 5),h(0.743 7) and Ne (4.4) ofthe population were higher than the part of other sheep breeds which were reported in the document andgenetic diversity were plenty which can be studied and analysised deeply.Secondly, by the micro-mark technology of PCR-SSCP, select the part of KAP1.1,kap1.3 and theextron KAP6.1which are the constitute protein of wool as the candidate gene. According to the research ofpolymorphism, the capability was found of it as the candidate gene of indirect selecting the fine trait ofwool. The result shows that focus W08667 of the high sulfur protein associated-protein(KAP1.1,KAP1.3)of keratin associated-protein family has significantly correlation with the fine of wool. Among thehigh-glycine-tyrsine keratin associated-protein, the BB genotype of W08667 has apparent relationship withthe fine of wool (P<0.05). The difference of BB genotype of locus W08667 and the original sequence hasnot change, and the single-nucleotide has not changed either. But the CC genotype has twosingle-nucleotide changed: the C of A+269 mutate into T,the G of A+168 mutate into A. By sequencing wefind that the AB genotype of W07399 there is one nucleotide mutation: C mutate into T. AA and ABgenotype of W08673 differ from the original sequence of W08673 : they unlike the above mentioned ,thesegene sequence was changed due to the insertion of nucleotide.
