A referencing flock comprsing eight paternal half-sib families totaling 374 individuals from Liangshan semi-fine wool sheep was utilised to analyze the allele frequency, polymorphism information content(PIC) and heterozygosity of nine microsatellite from 2nd chromosome of sheep genome.And the research based on the daughter design to locate the putative QTL affecting wool traits was carried out in the linkage framework-mapping of 2nd chromosome.The phenotypic traits comprising staple length(SLl),staple length at 18 months(SL2),staple length at 30 months(SL3),abdominal staple length at 18 months(ASLl),abdominal staple length at 30 months(ASL2),fleece weight at 18 months(FWl),fleece weight at 30 months(FW2) and wool fibre diameter at 30 months(WFD).The average number of alleles,average PIC and average heterozygosity of microsatellites were 7.56(from 4 to 10), 0.589(from 0.254 to 0.823) and 0.628(from 0.271 to 0.843) respectively.And all markers were high or middle polymorphic,which could substantially guarantee the reliability of the result of the research.The phenotypes of 281 progenies were analyzed by SAS6.12 statistical software.The simple statistical analysis demonstrated there was variation for every wool trait in certain extent,this ensured the location of QTL.The "CORR"consequence revealed the significant correlation between each wool trait.The"GLM" rusult demonstrated sire factor had great influence on SL1,SL2,ASL2 and FW2,meanwhile had significant influence on SL3 and ASL1.QTL analysis was performed by two software(QTL Express and WinQTLcart) which employ different methods(regression and LR test) respectively.There was a common putative QTL affecting WFD was detected.One was in family B3T96017,at 104cM between BMS1591 and OarCP0079 by QTL Express;the other was in family H1G4035,at 337.2cM between BMS2113 and OarFCB11 by WinQTLcart,and the distance between the QTL and OarFCB11 was 0.2cM.In addition,three putative QTLs that have great effect on FW2,SL1 and ASL1 were observed respectively by QTL Express.
