Studies On The System Of Tissue Culture And Plantlets Regeneration Of Langmu Series Hydrangea Macrophylla Ser

In this study, the stems from 'Hydrangea macrophylla ','Bolin', 'Lanxing' and 'Hudianxizhu' and were used as explants. By means of the orthogonal experiment design,analysis of variance and multiple comparisons,the factors including genotypes,basical media, explant type, the chroma of sugar ,different plant growth regulators and culture process were analyzed and the system of tissue culture and plantlet regeneration from axenic leaves and shoot was established.The results were as follows: (1) Initial culture'Hydrangea macrophylla'': 1/2MS+BA0.5mg/L+KT1.0mg/L+sugar30g/L, in which the germination rate reached 98.66% and the mean shoot length reached 4.22cm.'Bolin': 1/4MS+BA0.5mg/L+KT0.5mg/L+NAA0.5mg/L+ sugar 30g/L, in which the germination rate reached 100% and the mean shoot length reached 3.48cm.'Lanxing': MS+BA0.5mg/L+NAA0.1mg/L+ sugar 30g/L, in which the germination rate reached 99.9% and the mean shoot length reached 3.76cm.'Hudianxizhu': 1/4MS+BA0.5mg/L+KT0.5mg/L+NAA0.5mg/L+ sugar 30g/L, in which the germination rate reached 100% and the mean shoot length reached 3.12cm. (2)Multiplication culture'Hydrangea macrophylla Ser.': Two types for the multiplication happened.One was clump shoots genesis from no-buds proliferaten medium(type B),the other was clump shoots genesis from callus proliferaten medium(typeC).The optimal medium was MS+BA3.0mg/L +NAA 0.1mg/L +GA30.2mg/L +sugar 30g/L, in which the multiplication rate reached 25.08 and the multiplication of efficiency seedings reached 17.52.The better medium was MS+BA3.0mg/L+NAA0.1mg/L+GA30.2mg/L+ sugar 30g/L, in which One-step plantlet came true.'Bolin': type B;3/4MS+BA3.0mg/L+GA30.5mg/L+sugar30g/L, in which the multiplication rate reached 33.55 and the multiplication of efficiency seedings reached 15.29.'Lanxing': type C ;1/4MS+BA2.0mg/L+NAA0.1mg/L+GA30.5mg/L+ sugar 30g/L, in which the multiplication rate reached 12.33 and the multiplication of efficiency seedings reached 10.21.'Hudianxizhu': type C;1/4MS+BA3.0mg/L+NAA0.2mg/L+GA30.1mg/L+ sugar 30g/L, in which the multiplication rate reached 14.32 and the multiplication of efficiency seedings reachedl2.45.(3) Efficiency seeding culture'Hydrangea macrophylla':1/2MS+BA0.5mg/L+NAA0.05mg/L+GA31.0mg/L+sugar30g/L, in which the rate of efficiency seedings reached 97.79% and the rate of rooting reached 100%.'Bolin': 1/2MS+BA0.5mg/L+GA32.0mg/L+sugar20g/L,in which the rate of efficiency seedings reached 97.66% and the rate of rooting reached 100%.'Lanxing' : l/2MS+NAA0.1mg/L+GA31.0mg/L+sugar20g/L,in which the rate of efficiency seedings reached 96.53% and the rate of rooting reached 100%.'Hudianxizhu': 1/2MS+NAA0.2mg/L+GA32.0mg/L+sugar30g/L,in which the rate of efficiency seedings reached 98.63% and the rate of rooting reached 100%. (4)Rooting culture'Hydrangea macrophylla Ser.': 1/4MS+ sugar 10g/L, in which the rate of rooting reached 100% in 6.2 days.'Bolin': 1/2MS+IBA0.05mg/L+ sugar15g/L, in which the rate of rooting reached 100% in 4.7days.'Lanxing': 3/4MS+IBA0.1mg/L+ sugar20g/L, in which the rate of rooting reached 100% in 8.9 days.'Hudianxizhu': 3/4MS+NAA0.05mg/L+ sugar 15g/L, in which the rate of rooting reached 100% in 4.6 days.(5) TransplantingThe transplant medium in vermiculiteff, perlite and field soil by 2 :1:1 was the optimal for the four varieties. The rate of survival reached 'Hydrangea macrophylla' 86.07%, 'Bolin' 71.17%, 'Lanxing 75.83%' and 'Hudianxizhu' 65.03%.
