In the control, the leaves were treated by T0, and the leaves were cultured under the dark condition of 5,10,20,15 and 25d, respectively, and recorded as T1, T3, T2, T4 and T5 respectively. The relationship between regeneration frequency and POD activity,SOD activity CAT activity, MDA activity, soluble protein content and soluble surger was studied by H.macrophylla'Adri'Leaf.The results showed that the dark cultivation could not only contribute to POD activity grow, but also conduce to its activity remained at a high level. The SOD activity and soluble protein content could show the changes of leaf physio logical state, but could not reflect the effect o f dark cultivation on the regeneration frequency. In summary, dark treatment can help to incr ease POD activity and remain it at a high level.With the increase of dark incubation time, hydrangea leaf MDA content change is smaller, the activity can be reduced to about 10 mol/g;The content of soluble protein change is stable, but the experimental group volatile content of soluble protein is the longer the change.But the blade of dark train 15 d at the peak have a value of maximum,and the value can reach more than 30 mol/g.The change of soluble sugar content than the control group, There are two times the most resources.after 30 days earlier, the experimental group is a significant trend of high content of soluble sugar. CAT and soluble surger activity as indicators can explain the dark cultivation for cultivation effectiveness.
