The Toxicokinetics Studies On Haizaoyuhutang Decoction Etc. Three Prescriptions With Method Of Acute Mortality Of Mice

OBJECTIVE:To study the influence of compatibility of Sargassum and Radix glycyrrhizae and additional ingredients on the change of toxic dynamics characteristic of Haizaoyuhutang Decoction in mice. METHOD: three prescriptions studied including Haizaoyuhutang Decoction( â…  )and two new varieties of Haizaoyuhutang Decoction( â…¡ ,â…¢). according to the index of acute mortality of mice, three prescriptions were administered ip twice.we determined the remanet dosage in mice in different intervals of time after the first ip,then the apparent toxicokinetics parameters were worked out.RESULTS: The half lethal doses of â…  Decoction, â…¡ Decoction,and â…¢ Decoctin were 8661.637 , 6210.120 and 7564.844 mg/kg respectively. Intestine and liver tissue were injured in â…¡ Decoction;only intesine in I Decoction and â…¢ Decoctin. the first-order kinetics and one-compartment modle were noted on elimination of drug, after three prescriptions ip in mice.The apparent elimination rate constants were 0.0090, 0.0220 and 0.0261. The apparent elimination half-life periods were 77.4068, 31.4853 and 26.5316h.MRT were 11.628,29.785 and 10.738h respectively; AUC were 247.532, 74.033 and 92.047(mg/l)*h respectively.CONCLUSION Different proportion of Sargassum and Radix glycyrrhizae in a complex prescriptions result in changes of toxicity and pharmacokinetic characteristics of.Haizaoyuhutang Decoction. Additional ingredients on the basis of II Decoction can eliminate partly toxicity and pharmacokinetic characteristics of it.
