Herbal charcoals,one of the most characteristic processing methods of Chinese herbal medicines,were widely applied in the treatment of hemorrhagic disease,gastrointestinaltract disease and liver and kidney disease.However,it was uncertainly about the mechanism and its effect on other drug which combined with herbal charcoals.This study aims to put firm evidence for explain the mechanism of herbal charcoals by exploring the effect of Catnip charcoal on the Rhizoma Coptidis and Pulsatilla Decoction?four alkaloids in Rhizoma Coptidis including berberine,coptisine,palmatine,epiberberine;eight index component in Pulsatilla Decoction including berberine,coptisine,palmatine,epiberberine,esculetine,phellodendrine,aesculin and anemoside B4?.The adsorption test in vitro,0.5 g Catnip charcoal was suspended in 20 m L Rhizoma Coptidis aqueous extract or Pulsatilla Decoction aqueous extract with rotating.The content of each index component remained in the aqueous extract was measured at 5,10,15,30,60,120 min,respectively,with HPLC,then the Cumulative adsorption rate was calculated.Release test in vitro,1 g Catnip charcoal was suspended in 20 m L Rhizoma Coptidis aqueous extract or Pulsatilla Decoction aqueous extract with rotating,the precipitation was taken out to dry to constant weight after centrifugation.Then 0.5 g precipitation was suspended in 20 m L pure water with rotating.The content of each index component in pure water was measured at 5,10,15,30,60,120 min,respectively,with HPLC.The last experiment were set was the control.The animal experiment was implemented in two times,which 24 New masculinity Zealand rabbits were randomly divided into four groups.Groups in the first experiment were provided below,group A received aqueous extract of Rhizoma Coptidis?RCAE?,group B received RCAE plus CNC,group C received RCAE plus absorbent charcoal,group D received distilled water,respectively.Groups in the second experiment were provided below,group A received aqueous extract of Pulsatilla Decoction,group B received Pulsatilla Decoction plus CNC group,group C received Pulsatilla Decoction plus absorbent charcoal,group D received distilled water,Plasma samples from the rabbits were collected before and after administration.The concentrations of index component were measured by high-performance liquid chromatography?HPLC?after being of Catnip charcoal on four alkaloids in Rhizoma Coptidis were 33.46%,54.45%,22.37% and 41.44%,respectively.The cumulative adsorption rate of activated carbon on four alkaloid in Rhizoma Coptidis were 70.0%,82.5%,51.5% and 55.7%,respectively.The cumulative adsorption rate of Catnip charcoal on eight index components in Pulsatilla Decoction were 33.17%,54.32%,21.48%,42.01%,25.11%,32.11%,39.1% and 23.08%.The cumulative adsorption rate of activated carbon on eight index component in Pulsatilla Decoction were 33.17%,54.32%,21.48%,42.01%,25.11%,32.11%,39.1% and 23.08%.?2?Releasr experimentThe release concentration of Catnip charcoal on on four alkaloids in Rhizoma Coptidis were ranged from 2.73 to 7.23 ?g·m L-1 while the release concentration of activated carbon were ranged from 0.28 to 0.42 ?g·m L-1.The release concentration of Catnip charcoal on eight index components in Pulsatilla Decoction were ranged from 1.45 to 7.29 ?g/m L,respectively,while the release concentration of activated carbon were ranged from 0.28 to 0.42 ?g·m L-1.2 Animal experiment?1?Result of the effect of Catnip charcoal on the in vivo pharmacokinetics of the main alkaloids of Rhizoma Coptidis.The difference of area under the concentration–time curve of berberine was significant?P < 0.05?,and the highest one was Group B,Group C was the lowest one.The AUC of coptisine in group A was higher than group B,and the AUC in group B was higher than that of group C,the difference of coptisine AUC was significant?P < 0.05?.The AUC of palmatine in group A was significantly higher than that of group B was significantly higher than that of group A and group C?P < 0.05?,and the AUC in group A was significantly higher than that of group C?P < 0.05?.Conclusion:Carbonized Schizonepeta have sustained release effect on four alkaloid in Rhizoma Coptidis,including berberine,coptisine,palmatine and epiberberine and eight index component in Pulsatilla Decoction,including berberine,coptisine,palmatine,epiberberine,esculetine,phellodendrine,aesculin and anemoside B4.It also can increase bioavailability of berberine in Rhizoma Coptidis and berberine and phellodendrine and pulchinenoside B4 in Pulsatilla Decoction.
