Objective: To study the effects of dissolution contents of puerarin and Baicalin and Pharmacokinetics parameters and the bioavailability in rabbits of the components in ultramicro pulverization of Redix pueraria and Gegen Qinlian Decoction powder to provide useful reference and scientific testify for the clinical application of the Gegen Qinlian Decoction.Methods:1. The Redix pueraria was made into five different diameters powder. High performance liquid chromatography (HPLC) was applied to determine the contents of puerarin from five different diameters powder treated with water and 30% alcohol, to compare the dissolution differences.2. The Redix pueraria and Gegen Qinlian Decoction was made into ultramicro-pulverised powder, High performance liquid chromatography (HPLC) was applied to determine the contents of Puerain. The Radix Scutellaria and Gegen Qinlian Decoction was made into ultramicro-pulverised powder, High performance liquid chromatography (HPLC) was applied to determine the contents of Baicalin.3. The plasma concentration of pueraria and Baicalin was determined by HPLC in rabbits after they were administered with ultramicro-pulverised powder of Gegen Qinlian Decoction 13g·Kg-1·BW dosage by gastrogavage. The plasma concentration-time data of pueraria and baicalin was analyzed by Drug Analysis Software (DAS) .Results:1. Compared with different diameters of Redix pueraria, the contents of pueraria were extremely Significant different after treated with water; the group of ultramicro-pulverised powder was Significant different through 200 mesh after treated with alcohol , while the others were extremely Significant different.2. The contents of pueraria from ultramicro-pulverised powder of Redix pueraria and Gegen Qinlian Decoction were 3.20% and 3.05% after treated with alcohol; and the contents of Baicalin from ultramicro-pulverised powder of Radix Scutellaria and Gegen Qinlian Decoction were 9.69% and 3.94% after treated with alcohol3. The concentration-time curves of pueraria and baicalin were in accordance with two compartment model after the rabbits were administered with the ultramicro-pulverised powder of Gegen Qinlian Decoction 13g·Kg-1·BW dosage by gastrogavage respectively. The pharmacokinetics parameters of pueraria were as follows: t1/2a was 6.52±0.47h,t1/2β was 15.65±4.63h,AUC0-24 was 3.09±1.23 mg/L·h,AUC0-∞ was 3.73±1.56 mg/L·h,Tp was 1.0±0.78h,Cmax was 0.27±0.007 mg/L ; The pharmacokinetics parameters of baicalin were as follows: t1/2a 1.08±1.11h,t1/2β was 12.15±4.02h,AUC0-24 was 2.61±2.34 mg/L·h,AUC0-∞ was3.39±3.21 mg/L·h,Tp was 2.0h±1.23,Cmax was 0.21±0.007mg/L.Conclusion:1. The dissolution contents of puerarin were significantly increased with the decrease of the diameters of Redix pueraria after pulverized2. The contents of puerarin and baicalin were accorded with the pharmacopoeia, after redix pueraria and radix scutellaria made into ultramicro-pulverised powder.3. Pharmacokinetics study of puerarin and baicalin in ultramicro-pulverised powder of Gegen Qinlian Decoction in rabbits enriched the study of pharmacology of Chinese traditional medicine and supplied datas for further applied study of ultramicro-pulverised powder of Gegen Qinlian Decoction .
