Varietal identification is important to ensure the seed quality. In the present study, efficient identification methods for the main varieties of cowpea (Vigna unguiculata W.) and cauliflower (Brassica oleracea L.) in south of China, especially in zhejiang province, were investigated by using seven cowpea varieties (variety 1, Zhijiangtechang80;variety2, Zhijiangtechang90;variety3, Qiujiang512;variety4, Zhijiang228-2;variety5, Huajiang24;variety6, Yangjiang40;and variety7, Xingaochan8) and five cauliflower varieties (variety A, Longfenteda50tian;variety B, Yidaijinguang80tian;variety C, RuixuetedalOOtian;variety D, Qiuwang80tian;and variety E, Tezao50tian). The application of seed shape, seedling shape, rapid chemical identification and electrophoresis was performed, the main results were as follows.The results showed that there was no difference in seed color and luster among seven cowpea varieties, the seed length/thick ratio of five varieties was significant difference, the seed length/width ratio of one variety was identified from the others, so the six varieties could be identified by seed shape. The variety 3 could be identified by phenol and guaiacol test, the other varieties also could be classified into three groups. The variety 4 and 5 could be distinguished by length/width ratio of seedling leaves. In the present study, the lactic acid tank buffer (containing 15% sucrose and 0.05% methyl green) was selected as extraction solution, and seeds were put in water pot for 0.5h at 60 "C for protein extraction. By this method, the electrophoregrams gave abundance band diversity and could identified variety 1,3 and 5.There was a little difference in the colour of seed coat and embryo of cauliflower, but it was not obvious to identify varieties. The variety A, varieties B and E, varieties C and D could be distinguished by 10%NaOH solution treatment, the variety B also could be identified by HC1 and guaiacol test. By using the seedling shape identification, the variety C with purple colour was identified from the other varieties, the rest of varieties could be classified into two groups: variety A and B, variety D and E by leaf shape. The variety B, variety D and E could be identified by lactate-polyacrylamide gel electrophoresis by using protein complex extraction solution (containing 0.01 moL/L HC1, 6 mol/L urea and 0.05 % methyl green).It suggests that the protein electrophoresis could be used as a main method for cowpea variety identification, combining with other identification methods. The cauliflower varieties could be distinguished by the combination method of protein electrophoresis, the rapid chemical test and seedling shape.
