Identification researches of seed purity and genuineness for cowpea cultivars, Chinese cabbage cultivars, cauliflower cultivars and cucumber cultivars, which were extensively planted in Zhejiang province, were conducted using 14 RAPD arbitrary primers and 10 pairs of SSR molecular markers in the present paper. The main results achieved were as follows.1. DNA was extracted with celerity from the seeds of cowpea and cucumber according to a modified CTAB method. The optimal PCR system was established by muti-factor orthogonal experiment. 14 10-mer RAPD primers were used in identification of 7 cowpea cultivars, which proved the feasibility of the system established above. In addition, 4 (A7, A10, A9, B2) of the 7 RAPD primers can successfully distinguished all the 7 cowpea cultivars by being used together.2. The results showed that SSR molecular markers had not only the reliability, but also the high level of polymorphism and the eurytopicity in application. DNA from 60 cultivars of cowpea, Chinese cabbage, cauliflower and cucumber were amplified with 10 pairs of SSR primers. Among the 10 pairs of primers, the 5 pairs successfully distinguished all the 12 cowpea cultivars by being used together, the 4 pairs successfully distinguished all the 20 Chinese cabbage cultivars by being used together, the 2 pairs distinguished all the 10 cauliflower cultivars and the 3 pairs distinguished all the 18 cucumber cultivars.3. The present research also built the regression relationships between the purity results identified by SSR technology and the results by field test. The equations obtained can be used to predict the seed purity in the field.
