This research work is about isolation and identification of bioactive components andtheir dynamic distribution in aerial part of Stellera chamaejasme L.. The aim of thisresearch is to discover the bioactive components in the aerial part of the plant so that aerialpart of the plant can be used as a kind of resource of economic plant. In this work, theallelopathic effects of daphnin extracted from aerial part of the plant were also tested. Itwas designed to reveal the allelopathic substances existed in the aerial part of Stellerachamaejasme.Through solvent-extraction, silica gel Column Chromatography and determination ofUV, 1H-NMR, 13C-NMR, MS, X-ray et al., daphnoretin, daphnin and β-sitosterol wereseparated and identified from aerial part of Stellera chamaejasme. They all had antitumoractivity. Saccharose and another unidentified component were also isolated.The improved literature method of HPLC, which was used to measure the content ofdaphnoretin, was studied. HPLC condition of daphnoretin analysis was decided as follow:chromatogram column: Allteck C18 (250mm×4.6mm, 5μm);mobile phase: methanol /0.2% phosphoric acid solution (v/v) 45/55;detection wavelength: 305nm;columntemperature: 30℃;injection volume: 10μl. The retention time of daphnoretin was 7.3min.The HPLC instrument is LC-10ATvp from Shimadzu of JapanFor the aerial parts of Stellera chamaejasme from Daode zone, the highest content ofdaphnoretin was in May, viz.0.731mg/g DW (Dry weight). The data was 0.667mg/gDW inJuly and the lowest value was in August, viz. 0.294mg/gDW. Since September, daphnoretincontent raised and reached another highest value in October, viz. 0.662mg/gDW. Thedouble peaks of daphnoretin accumulation in aerial part of Stellera chamaejasme indicatedthat it could be harvested two times per year in Daode zone if daphnoretin was used asmain useful component. But the best season was in spring.For the samples from Changling zone, in May, daphnoretin content was the lowest,viz.0.533mg/gDW. In June daphnoretin content raised 0.687mg/gDW. The highest value ofdaphnoretin appeared in 15 July, viz.1.847mg/gDW. In August daphnoretin content reducedto 0.402mg/gDW. The best collection time in Changling zone was in middle of July.The quality of aerial part of Stellera chamaejasme from Daode zone was better thanChangling zone according to daphnoretin content.Allelopathic experimental results indicated that six concentration levels of daphnininhibited seeds germination of Vicia amoena and Melilotus offcinalis, but promotedgerminating of Lespedeza davurica. Different concentration of daphnin had dual effects tothe growth of radicle and embryo of the three species of legume plants. The promotingphenomenon happened in 0.05mg/ml level for three species of plants. When concentrationof daphnin exceeded 0.30mg/ml, the radicle and embryo growth of the three species ofplants were inhibited. The trend of allelopathic effects was the radicle and embryo growthof the three species of plants were improved in low concetration levels of daphnin andinhibited in high levels.
