A variety of extraction and separation methods were used to extract active components of Stellera chamaejasme L. in this study, and comparatively studied the influence of different extraction and isolation methods on total flavonoids content in Stellera chamaejasme L. First of all, the crude extract was extracted by using infusion methods and ultrasonic wave extraction method, respectively. Then, the polyamide adsorption method, acid-base precipitation method and solvent extraction method were used to further putifying the rude extracts. Eventually, the content of total flavonoids was calculated according to the rutin standard curve. Through reproducibility experiment, it has to approve the extract (SCAH) through infusion method and ultrasonic wave extraction method and reapplication of Solvent Extraction. Total flavonoids content of the extract (SCAH) is 24.78%. It not only has a high extraction rate of the active ingredient and a high utilization rate of the raw material, but also has lower cost, so it is suitable for a large number of extractions.Subsequently, immune stimulating effects of SCAH in mice were studied. Effect of SCAH on thymic and splenic indice were determined by weighting method; the phagocytosis of mononuclear phagocyte system was determined by using carbon particle clearance test; the contents of lysozyme in blood serum were measured by nephelometry respectively; Influence of SCAH on the content of IgM in blood serum was measured by blood serum hemolysin spectrophotometric method; The content of IgG,IL-2,TNF-É‘in blood serum was determined by ELISA method respectively. The results revealed that SCAH could increase the weights of thymic index of normal mice obviously and evidently enhance the content of IgG,IgM in blood serum, and can reinforce the phagocytosis of mononuclear phagocyte system and enhance the content of lysozyme in blood-serum of mice. It also can increase the content of IL-2, TNF-É‘in blood serum remarkably. Therefore, the SCAH can regulate immunological function of body.
