For dozens of years, with introducing of some fast-growing good mutton sheep breeds with high mutton production, natural specialties of local sheep breeds have been reducing and even lost because of unrealistic hybridization between introduced and local breeds, in addition to driving of market interest, continued worsening of ecological environment and other reasons. How to take effective and practical measures in supervision, evaluation, protection and utilization on the basis of natural value of local breeds, of current living conditions and commands of the market is of magnificent significance. This thesis provides scientific theoretical basis for making up practical countermeasures in protection and exploitation and precipitation by comprehensive evaluation of breed resources from biochemical inheritance according to sheep breeds' physiological features, geographical distribution, and local conditions such as climate, vegetation and precipitation. Long-Tail Han Sheep in Henan Province is a kind of good local breed with genetic stability, good adaptability and long history, and it has features such as extensive feed, coming into heat all the year round, multi-birth and multi-breeding, fast growing, high mutton production and high mutton quality. Since 1960s, beginning with sheep improvement by hybridization, pedigree breeding sheep have become domestic animals of imminent danger to be protected for reducing distribution and amount of livestock in hand just over 2000. Main problems to be solved involve too extensive feed management, lack of insemination station, lack of scientific guidance and effectivemanagement in breeding, and lack of research on interrelation between blood protein polymorphism and multi-lamb feature.The thesis provides scientific basis for breeding management, diagnosis of illnesses and evaluation of properties by survey of breeding nature, determination and relative analysis of physiological index and biochemical index to learn about Long-Tail Han Sheep in Henan Province in growth, breeding, nutrition and health from the angles of biochemical inheritance. Meanwhile, the thesis studies interrelation between multi-lamb feature and Hb and biochemical inheritance of Tf by examination of Hb and Tf with non-continuous vertical PAGE, and it provides theoretical basis for breeding of multi-lamb family and establishment and protection of gene bank from the angles of biochemical inheritance by calculation of gene frequence and genotype frequence to explore mechanism between Hb, Tf and multi-lamb feature of Long-Tail Han Sheep in Henan Province.Tests of blood physiological index of Long-Tail Han Sheep in Henan Province show that RBC, WBC, PVC, Hb, MCV, MCHC between male sheep and female sheep has no distinct difference. The sheep has great brittleness of red cell but little ESR. Ten blood biochemical indexes including serum total protein, serum albumin, serum globulin, serum total cholesterol, blood sugar, blood potassium, blood sodium, serum calcium, blood uric nitrogen and inosine are tested to 41 Long-Tail Han Sheep (females 19 and males 22), and the result shows that only serum cholesterol of the male is higher than that of the female ( P<0.01 ) , but other indexes between the male and the female have no distinct difference (P>0.01).Reference to Merion monotonous sheep, multi-state of Hb from Long-Tail Han Sheep in Henan Province is tested with non-continuous vertical PAGE, the result shows that Hb gene and HbBB genotype frequence of Long-Tail Han Sheep are 0.6945 and 0.5000 respectively, these two breeds" difference is distinct (p<0.05, p<0.01), and Hb genes of multi-lamb Long-Tail Han Sheep and single-lamb Merino monotocous sheep are all in superiority. But Hb gene frequence of Long-Tail Han Sheep is higher 26.27% than that of Merinomonotocous sheep, the difference is distinct after t test (p<0.01); HbBB genotype frequence of Long-Tail Han Sheep is higher 66.7% than that of Merino monotocous sheep, the difference is also distinct (P<0.01), this point may concern with its multi-lamb feature. It is possible to improve birth rate of progeny by choosing HbBB phenotype males and females to inseminate to improve lamb-breeding rate of future generations, so blood protein subtypes can be regarded as biochemical genetic marker to breed multi-lamb family of Long-Tail Han Sheep in Henan Province, to reduce gene frequence of Hb, to improve genotype frequence of HbBB, to set up multi-Iamb family step by step on the basis of special genes, and to improve frequence of good genes. Allelomorph Hb and Hb of multi-lamb Long-Tail Han Sheep and single-lamb Merino monotocous sheep are of Hardy-weinberg balance by X2 test. Reference to Merino monotocous sheep, polymorphism of Tf from Long-Tail Han Sheep is test, and it shows that among 36 bloods of Long-Tail Han Sheep five genes (Tf^Tf^Tf^Tf^Tf*) and eight phenotypes (TfAD, TfBB, TfBC, TfBD, TfCC, TfCD, TfCE, TfDD). Superiority of these five genes lists in turn Tf, Tf8. Tf0. Tf\ TfE, The highest lamb-breeding number of TfCD is two, others in turn are TfBC, TfDD, TfBD, TfAC, TfAB, TfAD, and the lowest lamb-breeding number is one. Different phenotype female sheep have different lamb-breeding number, but the difference is not significant. In the process of setting up multi-lamb family of Long-Tail Han Sheep, it worth considering to breeding males and females with TfCD, TfBC, and TfDD genotype to inseminate to improve gene frequence of Tf° and Tf8, and to improve lamb-breeding rate of future generations. Among thirty bloods of Merino monotocous sheep six genes (Tf\ Tf8. Tf0. Tf°. Tf* . Tf*4) and four phenotypes (TfBB, TfCC, TfAE, TfMD) are tested. Lamb-breeding number of different phenotype female sheep is one; superiority of these six genes lists in turn T^.T^.Ti^.tATi0 and Tf*. All allelomorphs at Tf loci of phenotype of multi-lamb Long-Tail Han Sheep and single-lamb Merino monotocous sheep are not of Hardy-weinberg balance by X test.
