In this paper, the tissue culture of Salvia miltiorrhiza and Glycyrrhiza uralensiswere studied.In the tissue culture of Salvia miltiorrhzia, the callus was induced from the leavesand adventitious root was induced from callus. The determination methods oftanshinoneⅡA and protocatechuic aldehyde were established. In addition, somefactors that influenced the tissue culture of adventitious root were maily studied.1.MS medium with a combination of 3% sucrose, IBA 2mg·L-1and KT0.2mg·L-1 was suitable for the induction and subculture of adventitious root.2.The biggest multiplication rate 8.0 was obtained at sucrose level of 60g·L-1;themaximum production of tanshinonⅡA 0.012mg·g-1 was achieved at sucrose level of20g·L-1;and the maximum production of protocatechuic aldehyde 0.22mg·g-1 atsucrose level of 30g·L-1.3.The biggest multiplication rate 5.1 was obtained at the pH value of 6.5;themaximum production of tanshinonⅡA 0.033mg·g-1 was achieved at pH value of 5.5and 6.0;and the maximum production of protocatechuic aldehyde 0.22mg·g-1 was gotat pH value of 5.8.4.The biggest multiplication rate 4.0 was achieved at inoculation rate of 2.5%.5.The biggest multiplication rate 4.17 was obtained with IBA 2.0mg·L-1;themaximum production of tanshinonⅡA 0.19mg·g-1was reached with a combination ofIBA 2.0mg·L-1 and KT 1mg·L-1;and the maximum production of protocatechuicaldehyde 0.34mg·g-1 was got with KT 0.5mg·L-1.6.The biggest multiplication rate 5.38 of adventitous root was obtained whileNH4+/NO3-was 1:4;the maximum accumulation contents of tanshinon Ⅱ A0.037mg·g-1 and protocatechuic aldehyde 0.34mg·g-1 were achieved when niratewas used as the sole N source.6.After exposed to US at 150W for 3 min, the biggest multiplication rate 2.1 ofadventitious root of S. miltiorrhiza was achieved;the maximum content of tanshinonⅡA 0.67mg·g-1 was reached at 300W for 3 min;and the maximum production ofprotocatechuic aldehyde 0.21mg·g-1 was abtained at 500W for 3min.In the tissue culture of Glycyrrhiza uralensis, the highest induction rate 98% ofcallus was obtained on MS medium with 2,4-D 2mg? L -1and KT 0.7mg? L-1 whenthe middle part of the root (0.5 cm) was taken as explant. In addition, the subculturetime for callus should be every 15days.
