Glycyrrhiza uralensis Fisch.,family Leguminosae,is a famous Chinese traditional drug, in which theflavonoids and glycyrrhizin were its main bioactive component. As natural resources can't fulfill require ofmedicine and other industries. Cultivation and tissue culture are considered to be hopeful way to alleviatethe crisis of natural resources. Tissue culture technic is a good method, it can control grow condition ofplants, accelerate pace of propagate,and extract active composition from cells.Glycyrrhiza uralensis Fisch.as material in the test. The course of callus culture,suspension culture andthe induction of hairy roots were studied in order to set up a whole tissue culture system, and so thatenhanced its secondary metabolite content, built the solid foundation for industrialization of the commercialproduction of effective component.The results are as follows:1.The optimal hormone combination for callus induction and proliferation were selected by the resultsof orthogonal test.According to the results of orthogonal test MS medium supplemented with 2,4-D(0.5mg/L),6-BA(1.5mg/L),NAA(1.5mg/L),KT(1.5mg/L)and IBA(1.5mg/L) was the most effective for callusinduction while MS medium supplemented with2,4-D(0.5mg/L),6-BA(0.2mg/L),NAA(0.2mg/L),KT(1.5mg/L)and IBA(1.0mg/L) was the most effectivefor callus proliferation.The total culture period was 30d,which the growth curve was assumed "S model".It is proved thegrowth of callus have the cyclical characteristic.2.The PAL activity in the cell treated with MJ and Asperillus were significantly promoted.A peakvalue is achieved in cycle 18d.3.The optimal culture conditons of cell suspension culture were selected by the results of OrthogonalTest.According to the results of orthogonal test MS medium supplemented with inculum size of7gFW/100mL,70g/Lsugar, pH=5.5(before sterilization),2.0mg/L2,4-D and 2.0mg/L NAA was the mosteffective for cell suspension culture.The total culture period was 21d, which the growth curve was assumed "S model".It is proved thegrowth of cell suspension culture have the cyclical characteristic.4.Hairy root of Glycyrrhiza uralensis Fisch.was transformed with Agrobacterium. However, thetransformation frequency of hairy root from explantation was low and the hairy root grow slowly.
