With medicinal research developed, a lot of consumers took more cautious attitude toward cosmetics containing chemical components. In recent years, more natural materials were used in cosmetic. The research and development on Cosmeceuticals would be the main direction of cosmetics industry.This subject was put forward at the background of aggravated trouble induced by hair loss disease people faced nowadays, which had a strong impact on people's mental outlooks and health. This study tried to find natural functional components from bamboo resource to develop new hair tonic and probe its mechanism on hair promotion. Several effective components from different pars of bamboo (include flavonoids of bamboo leaf, monomer homoorientin, triterpenes of bamboo bark, extract of bamboo node and extract of bamboo shoot) were researched on their hair promotion effects and mechanisms, by hair follicle organ culture and follicle cells culture in vitro and animal experiment in vivo. Meanwhile, the purification technology for flavonoids of bamboo leaf was also investigated. The objective of the study was to lie the foundation for development of hair tonic product containing bamboo extract.The main results and conclusions were as follows:(1) The influence on free hair follicle and follicle cells growth of effective components of bamboo The results demonstrated that the moderate dosage of flavonoids of bamboo leaf (0.005mg/mL) could promote the hair follicle growth significantly. Whereas low dosage (0.0005 mg/mL) showed suppression effect on free hair follicle growth and the high dosage (0.05 mg/mL) showed no obvious effect. Samples of three dosages retarded the period into logarithmic growth phase of dermal fibroblast (DF) compared with control group. Flavonoids of bamboo leaf of three dosages also inhibited keratinocyte cell (KC) and out root sheath cells (ORS) growth significantly.The moderate dosage of monomer homoorientin (0.0325 mg/mL) could accelerate free hair follicle growth and prolong follicle growth cycle but had no significance. The low dosage (0.00325 mg/mL) showed significant suppression effect on follicle growth and the high dosage (0.325 mg/mL) had no obvious effect. All of three dosages could promote DF growth significantly.. Both KC and ORS were however inhibited by three dosages of homoorientin.The extract of bamboo node indicated significant suppression effects on free hair follicle and three kinds of cells growth with high, moderate and low dosage.The triterpenes of bamboo bark (0.49 μg/mL) had no obvious effect on free hair follicle and ORS. But it could promote the DF growth and inhibit the KC growth significantly.(2) Preliminary study for the effect of extract of bamboo shoot on promoting animal hair in vivoThe results demonstrated that cavy and C57BL/6 mice could be both constructed as normal animal models to assess the hair promotion function. The study showed extract of bamboo shoot (EBS) could promote cavy hair growth, and also influenced the hair cycle of C57BL/6 mice. EBS had potential ability to promote animal hair growth.(3) The purification technology on separation of flavonoids of bamboo leafThe effect on separation of flavonoids of bamboo leaf by silica gel column chromatography was investigated , meanwhile, the results were compared with polyamide and AB-8 macroporous resin column chromatography by measuring several values of decolouring effect, yield, content of total flavonoids, HPLC chromatograph and effect of anti-free radical of DPPH ? , which could reflect different effects of three adsorbent materials on separation of flavonoids of bamboo leaf.The results demonstrated that reversed-phase column as polyamide and AB-8 macroporous resin column had shown better purification effect and selective ability on C-glycoside of flavonoids. But they couldn't separate flavonoids with polymerized pigments. The positive column as silica gel column, however showed better effect on separation of low polar and polar flavonoids, and had good decolouring effect.We take the conclusion that flavonoids of bamboo leafe, monomer homoorientin, triterpenes of bamboo bark and extract of bamboo node have different effects on free hair follicle, DF, KC and ORS cells growth. The function can be applied in different fields as promoting hair growth and occlusion of wound or inhibiting formation of psoriasis and cicatrix. Extract of bamboo shoot can promote cavy hair growth and influence the follicle cycle of C57BL/6 mice. EBS will be researched and developed as one kind of potential functional component for hair promotion. The feasibility of silica gel column chromatography used for separation and purification of flavonoids of bamboo leaf is accordance with demand of cosmeceutical quality. The effect on hair promotion and mechanism of effective components of bamboo will be researched in future.
