The efficient,stable plant regeneration system is a key to success for transformation. Factors affecting the high efficient regeneration were studied in vitro cultured shoots of widely used apple cultivaras Huangtaiping,jinghong and Shennong 2,including plant growth regulation,the length of darkness,polarity of leaf,basal media,type of explants,Ag NO3 and LH.hereby the high-frequency regeneration system of apple was established.The main results were as follows:1 The best results were obtained by culturing leaves adaxial side down on MS medium containing Sugar 30mg/L.The regeneration of tube shoots,the culture was with MS+TDZ2.0mg/L+NAA0.1 mg/L,MS+TDZ3.0mg/L+NAA0.1mg/L has the best result. Leaf of field shoots of Huangtaiping and Jinghong,the culture was with MS+BA5.0mg/L+ NAA0.05 mg/L,MS+BA8.0mg/L+NAA0.05mg/L has the best result; regenerative leavf also induce adventitious buds ,but the regeneration efficiency is not high. 2 For shennong 2, a period of darkness from 2 to 3 weeks was necessary to the high efficient regeneration system of adventition shoots.3 Leaf polarity best showed regenerated effciency of face of leaves downward was the best, and regenerated effciency was the best from the body leaf's base.4 Leaf, Petiole and Stem case of training the condition to be the same,the regenerated frequency of leaf was the highest.5 AgNO3 and LH had no acceleration to body leaf,but AgNO3 restrained growth of callus.6 With 1/2MS+IBA1.0mg/L+NAA0.1mg/L, it had the best result in rooting.7 Cytohistology showed the callus of leaves initiate the division of some vascular cylinder sheath cells; Development of adventitous buds belong to indirect methods.
