The cause of Acanthopanax Senticosus seed dormancy is studied in this article from the structure characteristic of seed shell (endocarp), the growth condition of the embryo, the endogene germination inhibitor. On the base of the system research on the seed dormancy reason, together with temperature-accelerated stratification method, the rule relevance of the embrionic growth and the material metabolism is discussed, so the seed dormancy mechanism is clarified initially.When fruit matures, the embryo is in the heart shape embryonic stage by microscope. It's structure is incomplete. The embryos ratio is only 2. 25%, without germination ability and need post- morphology maturation .The seed coats cornify, which can prevent the water and gas from exchanging, and result in the passive dormancy of the young embryos. But it is not the primary cause of the seed dormancy.By the use of system solvent separation method, three kinds of organic phase and water phase of the extract getting from the cosus pulp, the seed shell (endocarp) and the seed kernel are obtained. Among which, methanol phase inhibits the germination of cabbage seeds strongly. The turn: cosus pulp >the seed shell (endocarp) >the seed kernel. Therefore, methanol is a good kind of organic solvent to dissolve much inhibitors in some degree. After the methanol phase extracted from pulp is treated by pocket resin, 50% alcohol eluent inhibitory is strongest. It can be included that the existence of seed inhibitor is the primary cause of the seed dormancy.Use temperature-accelerated stratification method to relieve the Acanthopanax Senticosus seed dormancy. The seeds are stratified under lab condition. The average of high temperature is 19.36℃ (2~4 months), the embryo completes the differentiation of the seed leaf, the radicle and plumule in this period. And the average of low temperature is 4.11℃(5~6months), the embryo completes the physiological additional ripening in this period.During the process of temperature-accelerated stratification, the seed's breath speed, the soluble protein, the soluble sugar, the crude fat and the organic acid content are determined regularly including peroxide and catalase activity. The result of the determination indicates that crude fat content is reducing constantly, Sugar content increase in this process. Organic acid is middle product of these three kinds of nutrition substance. In the beginning, its content increased afterwards decreased sharply, which indicated metabolism is very active at low temperature. Metabolism speed is slow. The peroxide...
