To screen out the desirable genotypes and auxins for tissue culture, the effects of 3 auxins on the tissue culture from the immature embryos of 4 wheat genotypes (ie. Chuannong 16, Chuangyu 16, Chuangmai 42 and Liangmai 2) were investigated. The main results are described as following:1. The effects of 2,4-D, NAA, 2,4,5-T and the combination between 2,4-D and NAA on the tissue culture from immature embryos of wheat were investigated. It indicated that the addition of 2,4,5-T, NAA and the combination of 2,4-D and NAA in the media resulted in highly percentage of immature embryos germination and rootage. It was observed that 2,4-D was superior to 2,4,5-T and NAA in the frequency of immature embryos callus induction, germination and rootage. Optimum content of 2,4-D for the tissue culture of wheat immature embryos was 2.0-2.5 mg/L.2. The genotype is one of the key factors for the genetic transformation of wheat. Four genotypes (ie. Chuannong 16, Chuangyu 16, Chuangmai 42 and Liangmai 2) were used in this study. It was found that Chuangnong 16 was superior to other genotypes in the frequency of immature embryos callus induction, germination and rootage. Therefore, Chuannong 16 is a good donor genotype for wheat genetic transformation.3. Immature embryos are only available at one or two months in each year. To solve this problem, we put young spikes in 4 ℃ storage cupboards. At the same time the whole pretreatment is divided into seven time periods (7d). The result show that low temperature pretreatment was no obvious negative effect on the tissue culture from immature embryos of wheat.
