Study On The Tissue Culture Of Oat Immature Embryos And Regeneration

The study in this paper using the immature embryos of Husk Oats and Naked Oats affect callus induction by age of immature embryos, genotypes, mediums and 4℃processing;Studying the feasible subculture medium of Husk Oats (A. sativa .L.) and Naked Oats(Avena nuda L);studying the effect of Oats callus differentiation by mediums, genotypes, hormone combination and concentration. The results showed the best cultural media for different cultural stages were as follows:1)The feasible medium of Husk Oats: The callus inducing medium: MS+CH(500mg/L)+2,4-D(3.0mg/L)+NAA(1.0mg/L)+IAA(1.0mg/L); Subculture medium: MS+CH(500mg/L)+2,4-D(1.0mg/L)+6-BA(2.0mg/L); Differentiation medium: MS+CH(500mg/L)+NAA(0.3mg/L)+6-BA(1.0mg/L)2)The feasible medium of Naked Oats: The callus inducing medium: MS+CH(500mg/L)+2,4-D(2.0mg/L)+NAA(1.0mg/L)+IAA(1.0mg/L); Subculture medium: MS+CH(500mg/L)+2,4-D(1.0mg/L)+6-BA(2.0mg/L); Differentiation medium: MS+ CH(500mg/L)+NAA(0.5mg/L)+6-BA(1.0mg/L)3)The feasible rooting and strong medium of the two varieties: Rooting medium:1/2MS+CH(500mg/L)+NAA(0.5mg/L); Strong medium:1/2MS+CH(500mg/L)The effects of disinfection time, the period of drawing materials and the ways of placement on callus induction of oat immature Embryo were studied as well. The results showed that:disinfecting with 75% alcohol for 30 s, and then soaking with 0.1% corrosive sublimate can reach good effect of disinfection and survival percentage; The embryonic age 10-15d of immature embryos can form more callus blocks, and have the highest healing rate; In the ways of placement of oat immature embryo, upward placement of the shield could induce significantly higher rate of compact callus; downward placement could make young embryo germinate buds easily, and for the differentiation frequency of compact callus, no significant difference was observed between the two inoculation method.
