Indentifing HMW-GS of 15 common wheat recombinant inbred lines(RILS) with PCR technich and SDS-PAGE approach and determining protein content, hard, wet gluten content and SDS-sedimentation value by inframatic instrument, at the same time, analyse effects of HMW-GS on wheat quality traits. In order to know about HMW-GS diversity of Guizhou wheat varities, analyse HMW-GS of 138 wheat varieties with the SDS-PAGE approach, HMW-GS compositions, allelic variation , genetic diversity and frequencies of HMW glutenin subunits. The results were obtained as follows:1 Among those 15 RILS, there are 11 RILS ,which are stable. HMW-GS of 4 RILS that are unstable occurred seperation at Glu-A1 locus. According to the results of HMW-GS delection and principle that RIL which had same HMW-GS coming into being new line, there are up to 19 RILS at F8 genenation. At seedling stage of F8 gap, using PCR technich to delect HMW glutenin 1 or null at Glu-Al locus. The result showed that: there occurred a special band of about 1500 bp in lines which had 1 subunit, but not in lines which had null subunit. Conversely, RILS which had null subunit occurred a special band of about 920 bp,but PILS which had 1 subunit not occurred the band. The result of PCR identification and of the SDS-PAGE delection are unanimous, at the same time, PCR technich was great helpful to nanlyse good quality, such being simple, fast, accurate and scientific, so this method can be as served some theoretical basis for quality selection.2 The different HMW-GS and the different constitution types of HMW-GS diffentently effect the quality of wheat. There occurred 5 types of HMW-GS component in lines: (1, 7+9, 2+12),(null, 7+9, 2+12), (1, 7+9, 5+10), (null, 7+9, 5+10), (1, 20, 5+10) . The quality index of (null, 7+9, 2+12) is significantly superior to (1, 7+9, 2+12) ,and (1, 7+9, 5+10) is superior to (null, 7+9, 5+10) . As for as glutenin subunits at diferent Glu-1 loci are concend, at Glu-A1 locus, 1>null; Glu-B1 locus, 20>7+9; Glu-D1 locus, 5+10>2+12.3 with the analysis of HMW-GS of 138 wheat varieties (or lines) by SDS-PAGE, the results were showed that: â‘ Glu-1 loci had plentiful variant types and the HMW-GS compositions were abundant, and 15 subunit patterns and 33 subunit combinations were detected.
