The pure culture spawn was obtained from the sclerotia of wild Inontus obliquus bu tissue isolation from the area of mountain Changbai. The effect of pure culture and liquid culture of Inontus obliquus under different nutrition and environment condition were studied, the main influence factors were optimized by quadratic general rotational combination design with three factors and obtained the optimal pure culture and liquid culture mode of Inonotus obliquus mycelium; mutagenesis effect on Inonotus obliquus mycelium was also studied preliminarily.The results showed that the best pure cultivation mode for Inonotus obliquus is: the optimum nutrition substance and the best dosage is glucose26.428g/L, soybean power 1213.6g/L, CaSO43.44g/L,KH2PO41.5g/L,VB1 10mg/L;the optimum pH is 57, the optimum temperature is 25 30 ℃,complete darkness, appropriate gas permeation condiction. The optimal water content of Inontus obliguus mycelium by pure culture was 6065%,and the heaviest dry weight of mycelium is 19.2g/L. it is superior to other disposals .Taking the dry weight of mycelia as major evaluation index, at first to select the optimal liquid cultivation mode of Inonotus obliguus. mycelium, the optimal mode is: glucose 23.7 25.5g/L, peptone 1.21.4g/L,MgSO40.951.01g/L, KH2PO4 1.5g/L, VB1 10mg/L. While the optimum medium capacity, the culture temperature, the culture optimum pH, the spawning rate and the culture period were 140mL/250mL,25℃,7.0,170r/min,1012d,and the highest dry weight of mycelia was 0.1114g/L. It is superior to other disposals .Different UV irradiation time caused a certain inhibition action on average day growth rate of Inonotus obliquus mycelium, but the dry weight and content of polysaccharide of mycelium were increased evidently; the dry weight of mycelium of four minutes UV irradiation group was reached to 12.17g/L at the highest ;the content of polysaccharide of one minute irradiation groupwas reached to 2.39% at the highest.
