The purpose of this experimentation is to establish RP-HPLC chromatographic fingerprint of Alism a plantago-aquatica Linn by using RP-HPLC method, meanwhile study the total quality on different habitat sample, and the quality change of Alism a plantago-aquatica Linn at different growing-seedling stage, transplanting stage and sampling time and the selection and establishment of Alism a plantago-aquatica Linn GAP base by applying established RP-HPLC chromatographic fingerprint. The studying results showed as follows:1. The HPLC chromatographic fingerprinting of Alism a plantago-aquatica Linn, showing 26 characteristic peaks, was established from 42 lots of Alism a plantago-aquatica Linn samples. By comparison of the retention time and the on-line UV spectra of chemical standards, peak 16 and 22 were identified as 24-Acetyl Alisol A and 23-Acetyl Alisol B, respectively. The main diagnostic peaks were 16, 19, 22 and 24, and the ratios of peak area between 16, 19,22 and 24 were 1.064-1.153: 3.195-4.304: 1.000: 0.303-0.693.2. Samples from different habitats were of high similarity, most similarity ratio is at 0.94 upwards, which indicated that the total quality was very stable. But the samples purchased from medicinal materials market, which were machined, had evident difference from habitat samples. It had nothing more than 0.78 similarity, its ratio of examination for characteristic fingerprint was barely 0.75, and its ratio of difference was 1.42.3. When coming to the total quality, the total peak area of intercommunity peaks and 10 strong peaks on samples from Dou-Jiang weir city habitat was the best, secondly was the samples from Le- Shan city habitat, the samples from Peng-Shan County habitat were relatively inferior. The 23-Acetyl Alisol B content was the highest in Dou-Jiang weir city habitat samples, secondly was the Le- Shan city habitat samples; the 24-Acetyl Alisol A content was the highest in Le- Shan city habitat samples, Dou-Jiang weir city habitat samples was the second. The 23-Acetyl Alisol B and 24-Acetyl Alisol A contents were the lowest in Peng-Shan County habitat samples, moreover lower than 2 new producing areas. The 2 habitats of Alism a plantago-aquatica Linn—Dou-Jiang weir city habitat and Le-Shan city habitat should be the first producing area in the course of selection and establishment Alism a plantago-aquatica Linn GAP base.4. The peak area of intercommunity peaks and 10 strong peaks on samples gradually declined along with the postponement of growing-seedling stage. The total peak area of intercommunity peaks and 10 strong peaks in samples of 3 growing-seedling stages all had extremely remarked difference.
