Objective:To study the cell surface hydrophobicity and membrane permeability of Streptococcussanguis bacteriocin on Candida albicans(C.a) and Candida tropicalis(C.t). To furtherexplore inhibitory effect of Streptococcus sanguis bacteriocin on Candida.Methods:1. Recovery and extend standard strains Streptococcus ATCC10556,Streptococcus sanguisbacteriocin was extracted by low-temperature high-speed centrifugation, ultrasonic disruption.2.Used Adopt double dilution method to diluted different concentrations of Streptococcussanguis bacteriocin, and use Streptococcus sanguis bacteriocin on Candida albicans(C.a) and3.Using the method of microbial gelling hydrocarbons to make1g/L Streptococcus sanguisbacteriocin on Candida albicans(C.a) and Candida tropicalis(C.t) under37℃80rpmoscillating culture, measuring absorbance value of bacterial suspension and calculating thehrdrophobic of Candida albicans(C.a) and Candida tropicalis(C.t).4.Adding the Fluo-3AM to C.a and C.t, after12h, observing concentration of calciumioninto cell with Laser scanning confocal microscope.Results:1.The MIC of separate C.a and C.t is0.25g/L, The MIC of mixed C.a and C.t is0.5g/L.2.Under the influence of Sanguis Bacteriocin hrdrophobic of C.a and C.t reducingsignificantly, compared with the control group have significant difference(p<0.05).3.Discoid depression was appeared on surface of spore and hyphae after inhibition.Conclusion:1.Streptococcus sanguis bacteriocin could significantly inhibit C.albicans, C.tropicalis andtheir co-cultures.2.Streptococcus sanguis bacteriocin could significantly inhibit cell surface hrdrophobicityof separate and mixed cultured C.a and C.t.3.Streptococcus sanguis bacteriocin enhanced the calcium ion permeability of cellmembrane of C.a and C.t.
