Objective: The research was involved in discovering the mechanism of action when ergosterol of candida albicans suffering the influence of streptococcus sanguis bacteriocin of Streptococcus sanguis bacteriocin by detecting the change of spore and hyphal phase of candida albicansMethod:1.The standard strain of streptococcus sanguis ATCC 10556 were recovered and cultured and then the intracellular proteins of streptococcus sanguis(streptococcus sanguis bacteriocin)was extracted by centrifugation in low temperature,ultrasonic fragmentation and gradient salting out.2.The standard strain of candida albicans ATCC 10231 was recovered and oscillated cultured in Glucose-Saline-Biotin medium under 25? and 180 rpm for 48hours;3.Diluted the streptococcus sanguis bacteriocin to different concentrations with phosphate buffer saline,and then oscillated culture with spore and hyphal phase of candida albicans under 37? and 150 rpm.After 12 hours,we observed at times and get the the minimal inhibitory concentration(MIC)of streptococcus sanguis bacteriocin which worked on the spore and hyphal phase of candida albicans using the plate colony counting method.4.Using low temperature centrifugation to extracted the spore and hyphal phase of candida albicans respectively,which had streptococcus sanguis bacteriocin effected or not.We used the method of saponification to extract the cell of ergosterol and ultraviolet spectrophotometer to measure the absorbance of the cells which have co-culture with Streptococcus sanguis.Results:1.The MIC of hyphal phase of C.a is 0.25g/L,the MIC of spore phase of C.a is0.5g/L.2.Candida albicans spore cell membrane ergosterol content was 1.32±0.15%,and hyphal cell membrane ergosterol content was 0.63±0.01%,there was statistically significant difference(P<0.05).3.Mixed streptococcus sanguis bacteriocin with candida albicans to culture together:(1)In 12 hours,hyphal cell membrane ergosterol content significantly reduced,there was statistically significant difference between two groups(P<0.05);(2)In 24 hours,spore and hyphal cell membrane ergosterol content significantly reduced,there was statistically significant difference between two groups(P<0.05);(3)In 36 hours,hyphal cell membrane ergosterol content significantly reduced,there was statistically significant difference between two groups(P<0.05).Conclusion:1.Streptococcus sanguis bacteriocin showed significant inhibitory effect on spore and hyphae forms of candida albicans.And the inhibitory effect of streptococcus sanguis bacteriocin on the hyphal forms was stronger than the spore;2.The spore forms was higher than that of hyphal forms,which showed statistical significance(P<0.05);3.Streptococcus sanguis bacteriocin can decline the content of membrane ergosterol content of candida albicans,and it worked on hyphal stronger than spore.
