Objective:To investigate and determine the best formula of Tibetan medicine toothpaste combining toothpaste system with Tibetan medicine and study the anti-inflammatory effect and the antiseptic action of the Tibetan medicine toothpaste.To verify its anti-inflammatory effect and antiseptic action and make it have the function including cleansing and oral health care function preventing gingivitis, periodontitis and other oral diseases.Methods:First,to choose a reasonable and effective extraction process for Terninalia chabula Retz and Artemisia vestita Wal.Singe factor study and Orthogonal design were used to determine the optimized extraction process of Terninalia chabula Retz and Artemisia vestita Wal.The extraction rate and tannin content were used as the evaluating indicator to evaluate the extraction process of Terninalia chabula Retz.The extraction rate and oil extraction rate were used as the evaluating indicator to evaluate the extraction process of Artemisia vestita Wal.Second,to screen the formula of Tibetan medicine toothpaste.The friction agent, thickener, moisturizing factor and the dosage of water were the four main factors influencing the toothpaste molding.Formulating of recipe was used to determine the optimized formula of Tibetan medicine toothpaste using sensory properties as evaluating indicator. Using xylene induced mouse ear swelling model,the ear swelling volume was measured to analyse the anti-inflammatory effect of Tibetan midicine toothpaste.The oral pathogens selected were Streptococcus parasanguinis(ATCC 15909),Streptococcus mutans (ATCC25175),Actinomyces viscosus(ATCC15987),Lactobacillus acidophilus(ATCC4356).minimal nhibitory concentration and minimal bactericidal concentration were determined by liquid dilution method to evaluate the antiseptic action of Tibetan medicine toothpaste. Last,to establish a RP-HPLC method for the determination of gallic acid in Tibetan medicine toothpaste.Results:The optimized extraction process of Terninalia chabula Retz is as follows:8-fold 30% ethanol solution added,soaked for half hour,extracted for half hour with two time.The optimized extraction process for Essential Oil of Artemisia vestita Wall is as follows:18-fold water added, soaked for 1 hour, extracted for 5 hours.The optimized formula of Tibetan medicine toothpaste using Silicon dioxide as abrasive is Tibetan medicine 3.3 g,glycerin 25 g,70%sorbitol 40 g,Silicon dioxide 22 g,non-ionic guar gum 0.7 g,oleum menthae 1.0 g,sodium dodecyl sulfate(K12) 2.2 g,ethylparaben 0.1 g,propylparaben 0.05 g,saccharin 0.35 g and water right amount.The optimized formula of Tibetan medicine toothpaste using calcium hydrophosphate as abrasive is Tibetan medicine 3.3 g,glycerin 10 g,70%sorbitol 16 g, calcium hydrophosphate 52 g, non-ionic guar gum 0.6 g, CMC-Na 0.4 g,oleum menthae 1.0 g,sodium dodecyl sulfate(K12) 2.4 g,ethylparaben 0.1 g,propylparaben 0.05 g,saccharin 0.35 g and water right amount.The preliminary evaluation of the anti-inflammatory effect of Tibetan medicine toothpaste indicated that Tibetan medicine toothpaste markedly supressed the ear swelling induced by dimethylbenzene.The results of bacteriostatic action of Tibetan medicine toothpaste indicated that the Tibetan medicine toothpaste was not bacteriostatic effect within the scope of Tibetan medicine extract added to the medicine concentration.But considering the high pressure steam sterilization damage to essential oil from Artemisia vestita Wall,antibacterial effect of essential oil from Artemisia vestita Wall was separately studied.The result indicated that essential oil from Artemisia vestita Wall in Tibet have good antibacterial effects, for Streptococcus parasanguinis, the MBC is 0.734 mg·ml-1,for Actinomyces viscosus, the MBC is 0.734 mg·ml-1, for Streptococcus utanscillus, the MBC is 0.092 mg·ml-1, for Lactobacillus acidophilus, the MBC is 0.0223 mg·ml-1,for staphylococcus aureus, the MBC is 2.938 mg·ml-1.The essential oil from Artemisia vestita Wall added to toothpaste had reached the minimum bacteriostasis concentration,which indicated that the Tibetan medicine toothpaste has good antibacterial effect.Essential oil from Artemisia vestita Wall was as the main effective components of the bacteriostatic action.The study using high performance liquid chromatography to determine gallic acid in toothpaste show that the methanol-0.1% triethylamine- 0.1% phosphoric acid solution was as mobile phase with gradient elution.The method can make the gallic acid peak achieve baseline separation.Determination of gallic acid in Tibetan medicine toothpaste was average content of 0.802 mg·g-1.Conclusion:Tibetan medicine toothpaste had stable property. Its paste has exquisite and light,cool and refreshing smell,moderate viscosity,and has good anti-inflammatory and antibacterial effect. Tibetan medicine toothpaste had a broad market prospect.
