| Objective:Discussion the possible mechanism of action about of electroacupunct-ure Siguan acupoint to the post-ischemic stroke depression rats by observing the effect on the content of NE,DA and5-HT in the brain tissues of the prefrontal cortex and hypothalamus.Methods:The70rats were divided into five groups randomly by three times,res-pectively as the normal group,stroke group,PSD group,Siguan group, and fluoxeti-ne group.Except normal group,the rats of other groups were used to establish left side of the MCAO models by modified Koizumi filament method.PSD modelrats were prepared by living alone in combination with mild stimulation bychronic u-npredictableafter stroke model success7days.After the completion of successful modeling and grouping,normal group, stroke group, and PSD group were not tre-ated.The Siguan group were received the treatment of electroacupuncture Siguan acupoint,20minutes/time/day,and the fluoxetine group were gastrically perfused with fluoxetine with the dosage of2.33mg/(kg.d),both continuous treatment for21days.The all rats were received nerve function evaluation,sugar consumption e-xperiment and openfield test at different time points of unity.After21days,the contents of NE,DA and5-HT in the prefrontal cortex and hypothalamus of rats were measured by RP-HPLC-fluorescentmethod.Results:1.The neurological severity score of PSD rats were significantly increa-sed compared than normal group and stroke group (P<0.01),and the duration of the horizontal sticks were obviously reduced (P<0.01).However,electroacupunctu-re Siguan acupoint and fluoxetine gavage can markedly reduced the neurological severity score and increased the duration of the horizontal sticks (P<0.01), but the effect of Siguan group is superior to fluoxetine group (P<0.01),and Siguan group takes effect faster.2.The sugar consumption ratio, crawl lattice number an derect frequency of PSD rats were obviously reduced than the normalgroup and stroke group (P<0.01).However,electroacupuncture Siguan acupoint and fluoxeti-ne gavage can markedly increased the sugar consumption ratio,crawl lattice num-ber and erect frequency (P<0.01),the effect of the two groups is quite in incre-ase the sugar consumption ratio (P>0.05),and the effect of Siguan group is ob-viously superior to fluoxetine group in improving crawl lattice numberand erect number,but Siguan group takes effect faster (P<0.01).3.The contents of NE,5-HT in prefrontal cortex and the contents of NE,DA and5-HT in hypothalamus of PSD rats were significantly lower than the normal group and stroke group (P <0.01).However,electroacupuncture Siguan acupoint can obviously increase the contents of each at the same time (P<0.01).But, fluoxetine only can significant-ly increase the contents of5-HT in two brain tissues (P<0.01),and there were no difference between the two groups at it (P>0.05),but can’t return tonormal level.Conclusions:The trement method of electroacupuncture Siguan acupoint can ob-viously improve the neurological severity score and the indicatorsof associated w-ith depression of PSD rat model,can be effective treatment the PSD,and the effe-ct of Siguan group is obviously superior to fluoxetine group.Its mechanism may be related with the increased contents of NE,5-HT in prefrontal cotex and the c-ontents of NE,DA and5-HT in hypothalamus at the same time. |
