Objective: In order to set up the posterior stroke depressive rat model. To estimate the treatment of YuLeShu on the neurological functional recovery of stroke rat . To investigate the influence of YuLeShu on the depressive behavior and cerebral monoamine neurotransmitters of PSD rat model. Furthermore, to probe the anti-depressive mechanism of YuLeShu.Methods: Middle cerebral artery is occluded by advanced Koizumi's method to set up focal cerebral ischemic rat model.On the basis, further isolated-living condition and moderate unpredictable stress are applied successively to set up PSD rat model . The neurological functional recovery of stroke rat is assessed by Longa 5 grades and horizontal bar test.Sucrose solution consumption test, open-field test and passive avoid test are used to estimate the change of interest, the level of spontaneous motor activities and investigative behavior of PSD rat model.Fluorophotometry is applied to detect the monoamine neurotransmitters(NE,5-HT and DA) of the double frontal-parietal cortexes and brain stem. Investigating the action of YuLeShu on the neurological functional recovery of stroke rat. Evaluating the efficiency of YuLeShu on the depressive behavioral tests and cerebral monoamine neurotransmitters of PSD rat model.Results: The PSD rat model manifests significant changes: neurological functional impairment, weight loss, the decrease of sucrose solution consumption, the droping of open-field behaviors, the deficiency of passive avoid test, the droping of cerebral monoamine neurotransmitters. Low-dose, middle-dose and high-dose groups of YuLeShu promote the neurological functional recovery of stroke rat. High-dose group of YuLeShu augments the consumption of sucrose solution,besides, improves the open-field behaviors of PSD rat significantly. Middle-dose and high-dose groups of YuLeShu meliorate the deficiency of passive avoid test and increase cerebral NE and 5-HT concentration of PSD rat significantly.Conclusions:On the basis of focal cerebral ischemic rat model, isolated-living condition and moderate unpredictable stress are applied successively to set up suitable PSD rat model . YuLeShu can promote the. neurological functional recovery of stroke rat.It can also ameliorate the depressive behaviors and cerebral monoamine neurotransmitters concentration of PSD rat model . The anti-depressive mechanism of YuLeShu manily is due to increases the cerebral NE and 5-HT concentration and the systemic protection against cerebral ischemia.
