Objective:To observe the effects of Liu Wei Dihuang Decoction on insulin receptor substrate2and Phosphatidylinositol-3-kinase in liver in type2diabetic rats. From the molecular and genetics level, exploring the mechanism of Liu Wei Dihuang Decoction intervented in type2diabetes and improved insulin resistance. Providing reference for the clinical laboratory with type2diabetes to find new therapeutic targets.Methods:On the basis of our previous animal experiments, taking fresh liver tissue of rats from each group; And FBG levels were tested, INS levels were detected by ELLSA method;Real-time fluorescent quantitative PCR (RT-PCR) method was used to determine mRNA expression of insulin receptor substrate2(IRS2), phosphatidylinositol3-kinase(PI3K) in liver; IRS2> PI3K protein expression level were determined by Western Blot.Results:1.Compared with the model group, Liu Wei Di Huang Decoction group could reduce the FBG and INS levels in type2diabetic rats.(P<0.05)2. In Liu Wei Dihuang Decoction group, the expressions of IRS2and PI3Kp110αin the rats’ liver were significantly higher than the model group (P<0.05)3. Compared with the model group, the protein expressions of IRS2and PI3Kp85in the liver were significantly increased In Liu Wei Dihuang Decoction group.Conclusion:Liu Wei Di Huang Decoction could increase the mRNA and protein expression of IRS2and PI3K which were important factors in type2diabetes model rats’liver insulin signaling.It may be the internal mechanism that Liuweidihuang Decoction have been able to intervene in type2diabetes and improve insulin resistance. By regulating the expressions of IRS2and PI3K at upstream to affect the insulin metabolism and biological of downstream in PI3K/Akt pathway. Thus intervened to sugar metabolic processes in insulin resistance type2diabetes rats and biological effects of hepatic insulin resistance.
