Objective To observe the influence of Zhibai Dihuang Decoction (Zhibai Dihuang Decoction ZDD)on rortine semen indicators and expression of spermatogenic cell apoptosis factors Cyt-c, AIF in Ureaplasma urealyticum (Ureaplasma urealyticum UU) infected rats, then to explore the pathological mechanism of UU infection cause infertility and the intervention effect of ZDD.Methods45out of60SD rats from4to5mouth old were randomly selected and made into the UU infected animal model by bladder inoculation. The rest15rats were simultaneously received normal saline injection as the normal control group. On the7th day after inoculation, culture method was used to detect UU in the abstracted testis puncture fluid.which was for judgment of modeling success. After success of modeling, the UU infected model rats were then randomly divided into model group, azithromycin group (at the dose of0.105mg/kg. d, which means1ml/d), ZDP Group (at the dose of0.105mg/kg. d, which means lml/d). Equal volume of normal saline was given to the rats in normal group and model group. The rats of above groups were fed by intragastric administration,1times a day, chronically administered for21d. Rats were sacrificed and the epididymides tissue was extracted. Sperm cells were isolated by mechanical separation. Sperm motion parameters were detected by color sperm dynamic detection system. UU culture results and spermatogenic cell apoptosis rate were detected. ELISA method was used for detection of the expression level of spermatogenic cell apoptosis factors Cyt-c and AIF.Results When compared with normal group, the levels of A-grade sperm, B-grade sperm, linear velocity, average velocity were significantly lower but UU culture positive rate and spermatogenic cell apoptosis rate was significantly increased in model group. There were statistically significant differences in expression levels of spermatogenic cell apoptosis factors Cyt-c and AIF (p<0.01) between the above groups. After treatment, UU culture positive rate was decreased in both Traditional Chinese medicine(Traditional Chinese medicine TCM) group and Western medicine group when compared with model group and there was statistical differences (p<0.05)between them, but it showed no statistical significance (P>0.05)between TCM group and Western medicine group. The levels of A-grade sperm, B-grade sperm, linear velocity, curvilinear velocity, average velocity were improved both in the two groups and there were statistical differences(p<0.05, p<0.01) when the above two groups were compared with model group.While both the two groups can’t improve the levels of linear motion. TCM group was better in improving the levels of A-grade sperm, B-grade sperm, curvilinear velocity, average velocity than western medicine group and the difference was statistically significant (P<0.05, P<0.01). The expression levels of Cyt-c and AIF were also significantly lower in the two groups (P<0.01), but the difference was no statistically significant(P>0.05).Conclusion1.ZDD can effectively inhibit the growth, reproduction of UU then has the effect of anti-UU.2. UU infection can lead to spermatogenic cells apoptosis quantities of rats increase, which reflect the decline in semen quality is due to excessive apoptosis of spermatogenic cells caused by UU infection. Sperm quality decline can further lead to occurrence of infertility.3. The enhancement of expression levels of Cyt-c and AIF in UU infected rats’spermatogenic cells shows that it is an important factor to promote apoptosis. And apoptosis is of multi-approach.4. ZDD can decrease the expression levels of Cyt-c and AIF, which illustrat that one of the mechanisms in describing ZDD in the treatment of UU infected rats and improving their semen quality is regulating the expression of Caspase dependent way cells apoptosis factor Cyt-c and non Caspase dependent pathway cells apoptosis factor AIF.Thus ZDD against apoptosis in multi-path.
