Purpose:To observe the effect of Modified by removing water effect on S180ascites of mice before and after medication abdominal circumference change, theconcentration of the ionconcentration and Na+、 K+、NO in the blood, and theeffect on S180ascites mouse Peritoneal lymphatic stomata diameter, Peritoneallymphatic stomata perimeters,peritoneal lymphatic stomata area, peritoneallymphatic stomata density, and to investigate the Modified by removing watermechanism for the treatment of malignant peritoneal effusion.Material and method:1.Experimental animal and cell lines50Kunming mice, male and female, body weight (20±2) g,6to8weeks ofage, were purchased from experimental animal center of Liaoning University ofTraditional Chinese Medicine. S180sarcoma cell line (ascites), purchased fromLiaoning University of Traditional Chinese Medicine biotechnology laboratory,conventional culture in vivo passage, regular aseptic ascites, centrifugationsupernatant was added into the freezing medium gradient cooling,-80℃deepfreezer frozen.2.Experimental methods(1) The establishment of S180ascites mouse modelUnder aseptic conditions, has made10S180ascites of Mice Ascites prepared,the preparation of a S180ascites tumor cell suspension (tumor cell survival rate>95%, cell concentration of1×106/mL) under aseptic condition injection to32healthy mice were randomly selected, each mouse was injected into the0.2mL,7days after the32S180ascites mice all successful modeling, all rats survived,ascites homogeneous good.(2) Animal grouping and drug delivery method Randomly,40mice without8mice injected with S180ascites tumor cellsuspension into the blank group, the other has made32S180ascites of mice wererandomly divided into4groups. The control group, respectively, the westernmedicine group (Shun Bo), the Chinese medicine group (modified by removing waterparty), the combined group (Shun Bo+Modified by removing water), with8ratsin each group, a total of5groups.Gavage dose according to human and rat dose conversion, formula was usedto calculate dose. Blank group: daily with normal saline0.5mL/only orally;control group: daily with normal saline0.5mL/gavage; western medicine group(Shun Bo): daily with normal saline0.5mL/gavage; Chinese medicine group(Modified by removing water): daily to Modified by removing water0.5mL/gavage;combination group (Shun Bo+Modified by removing water): daily to Modified byremoving water0.5mL/gavage. Experiment first day and seventh day, westernmedicine group (Shun Bo), combination group (Shun Bo+Modified by removing water)give the configured Shun Bo0.2ml/intraperitoneal injection (10mg Shun Bodissolved in17.4ml saline); blank group, control group, Chinese medicine group(Modified by removing water) with normal saline0.2ml/rats injection. Compared with theblank group, control group, western medicine group (Shun Bo), Chinese medicinegroup (modified by removing water) and combination group (Shun Bo+Modifiedby removing water) circumference difference increased obviously, the differencewas statistically significant (P<0.05); compared with the control group, westernmedicine group (Shun Bo), Chinese medicine group (modified by removing waterparty), combination group (Shun Bo+Modified by removing water) circumferencedifference decreased significantly, the difference was statisticallysignificant (P<0.05); and the western medicine group (Shun Bo), combinationgroup (Shun Bo+Modified by removing water) circumference difference decreases,the difference was statistically significant (P<0.05); and the Chinese medicinegroup (modified removing water), combination group (Shun Bo+Modified byremoving water) circumference difference decreases, the difference wasstatistically significant (P<0.05). The combined group (Shun Bo+Modified byremoving water) is more obvious on mice abdominal perimeter control.2.Modified by removing water influences on the changes of Na+、K+concentrationin S180ascites in mice bloodExperimental results show that: compared with the blank group, control group,western medicine group (Shun Bo), Chinese medicine group (modified by removingwater) and combination group (Shun Bo+Modified by removing water) Na+、K+concentration increased significantly, the difference was statisticallysignificant (P<0.05); compared with the control group, western medicine group(Shun Bo), traditional Chinese medicine group (modified by removing water),combination group (Shun Bo+Modified by removing water) Na+、K+concentrationdecreased, the difference was statistically significant (P<0.05); and thewestern medicine group (Shun Bo), Chinese medicine group (modified by removingwater) K+concentration, the difference was statistically significant (P<0.05);and Chinese traditional medicine group (modified by removing water),combination group (Shun Bo+Modified by removing water) K+concentration decreased, the difference was statistically significant (P<0.05). The combinedgroup (Shun Bo+Modified by removing water) is more obvious for Na+、K+concentration in mice blood decreased.3.Modified by removing water effect on the change of NO concentration in S180ascites in mice bloodExperimental results show that: compared with the blank group, control group,western medicine group (Shun Bo), Chinese medicine group (modified by removingwater) and combination group (Shun Bo+Modified by removing water) NOconcentrations significantly decreased, the difference was statisticallysignificant (P<0.05); compared with the control group, western medicine group(Shun Bo), Chinese medicine group (modified by removing water), combinationgroup (Shun Bo+Modified by removing water) NO concentration increased, thedifference was statistically significant (P<0.05); and the western medicinegroup (Shun Bo), Chinese medicine group (modified by removing water),combination group (Shun Bo+Modified by removing water) NO concentration ishigh, there is statistically significant differences (P<0.05). The combinedgroup (Shun Bo+Modified by removing water) on blood concentrations of NOincreased more obviously.4. Modified by removing water effect on the change of peritoneal lymphaticstomata perimeter, area and density concentration in S180ascites mouseExperimental results show that: compared with the blank group, control group,western medicine group (Shun Bo), Chinese medicine group (modified by removingwater) and combination group (Shun Bo+Modified by removing water) of theperimeter, area and density increased significantly, the difference wasstatistically significant (P<0.05); compared with the control group, westernmedicine group (Shun Bo), Chinese medicine group (modified by removing water),combination group (Shun Bo+Modified by removing water) area and thedistribution density of the perimeter, increases, the difference wasstatistically significant (P<0.05); and the western medicine group (Shun Bo),Chinese medicine group (modified by removing water), combination group (Shun Bo+Modified by removing water) the area and perimeterdistribution density,increases, the difference was statistically significant (P<0.05). The combinedgroup (Shun Bo+Modified by removing water) perimeter, area and density ofstomata increased more significantly in mice.Conclusion:1.Modified by removing water on S180ascites in mice before and aftermedication changes in abdominal circumference decreased significantly, Modifiedby removing water eliminate abdominal water diuresis effect, has good effecton relieving the symptoms of mouse ascites.2.Modified by removing water can reduce the concentration of S180ascitesin mice blood Na+, K+, reduce water sodium retention, to eliminate the effectof ascites.3.Modified by removing water can increase the concentration of S180in bloodof Mice Ascites NO, so as to promote lymph peritoneal cavity open, to eliminatethe purpose of ascites.4.Modified by removing water can significantly promote S180ascites mouseperitoneal lymphatic stomata opening, the peritoneal lymphatic stomata area,perimeter increases, the distribution density of the lymphatic stomataincreased.
