[Objective]To investigate the association between a 45bp insertion/deletion(Ins/Del) polymorphism in the 3’-untranslating regin(3’-UTR) of tis exon 8 in uncoupling protein 2(UCP2) gene and type 2 diabetic retinopathy in Yunnan Province.[Methods] In this study, choosed the patients with type 2 diabetes mellitus (T2DM) as the research objects,who were hospitalized in Yunnan Province Second People’s Medical Endocrinologist and Ophthalmology between June 2012 and October 2014.The type 2 diabetic patients will be divided into non-retinopathy group (NDR group),non-proliferating retinopathy group (NPDR group)and proliferating retinopathy group (PDR group),in addition,in addition,Taking regular physical examination for normal control group(N group)in our hospital medical center at the same time. Record the clinical data,laboratory test of the biochemical indexes were made.The. subjects of UCP2 3’-UTR I/D were detected by using polymerase chain reaction (PCR),then sequencing identification,recording genotype and allele distributions, statistical analysis was made.[Results] 1.The allele frequency(P=0.590; OR=1.13[0.73-1.75]and the genotype distribution (P=0.620)were no significant differences between N group and T2DM group.2.There were significant differences in the allele frequency (P=0.024; OR=1.66[1.07-2.59]) and genotype distribution DD(57%、ID(42%),II(1%) (P=0.030) between NDR group and DR group, the risk DD. DI genotype distribution at UCP2 3’-UTR I/D were colsely assiciated with DR in Yunnan Province.3. Binary Logistic regression analysis:the risk DD、DI genotype distribution at UCP2 3’-UTR I/D gene were colsely assiciated with DR in Yunnan Province.4. Using relevant factors and Logistic regression analysis:the cardiovascular diseases and duration were positively correlated with diabetic retinopathy (p<0.05), who were the risk factors of DR.[Conclusion] 1. There is no significant difference in UCP2 3’-UTR I/D genotype distribution and allele frequency between N group and T2DM group.2.It is significant differences in UCP2 3’-UTR I/D genotype distribution and allele frequency between NDR group and DR group.3. Between normal group and the DR grcmp,the allele frequency A(51%),G(49%)(P=0.047;95%CI]= 1.45[1.00-2.0)and genotype distribution AA(23%)、AG 56%)、GG(21%) have statistically significant(P=0.003).3. Between NDR group and the NPDRgro哗,the allele frequency(P=0.150; OR95%CI]= 1.38[0.89-2.14])was significant difference the genotype distribution AA( 23%)、AG(56%)、GG2l%)have statistically significant(P=0.003).4. There were significant differences in the genotype distribution AA(23%)、AG(56%)、GG 18(21%) have statisticallysigniificant(P=0.030) between NDR group and PDR group,the allele frequestion(P=0.051; OR95%CI]=1.50(1.00-2.24) was no significant difference.5.Binaiy Logistic inegression analysis confirmed:the risk genotype distribution GG at UCP2-866G/A gene(rs65%9366 was closely associated with DR in Yunnan Province.[Conclusion]1.There is no significant difference i打 UCP2-866G/A genotype distribution and allele frequency between normal group and T2 DM group.2. There is significant difference in UCP2-866G/A genotype distribution in the NDR group and DR group.3Jt indicates that carrying GG genotypes increase the risk of DR in Yunnan Province.
