Purpose:This thesis aims to explore the functional mechanism of Qingzhi Tongmai Granules in preventing atherosclerosis(AS)through observation of the influence that Qingzhi Tongmai Granules exerts on the expression level of interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)in blood lipid metabolism as well as serum of the experimental rabbits with atherosclerosis.Material and method:1 Select fifty New Zealand rabbits and make them to become animal models with hyperlipoidemia by feeding them high-fat diet. Divide these fifty rabbits into five groups randomly with each e group ten rabbits:(1)blank control group;(2)hyperlipoidemia control group;(3)Simvastatin positive control group;(4) Qingzhi Tongmai granules low dose group;(5)Qingzhi Tongmai granules high dose group.2 At the end of 13-week experiment,(1)Detect contents of total cholesterol(TC), triglyceride(TG),low density lipoprotein cholesterin(LDL-C) and high density lipoprotein cholesterin(HDL-C)in blood serum with use of the automatic biochemistry analyzer;(2)Use enzyme linked immunosorbent assay(ELISA)to measure interleukin-6(IL-6)and expression level of tumor necrosis factor-α(TNF-α)in serum.Results:1 Results from each group of rabbits blood lipid show that:compared with the blank control group,contents of TC,TG and LDL-C has an incredible increase(p<0.01)and HDL-C has an obvious decrease(p<0.01).Each dose group of Simvastatin tablets and Qingzhi Tongmai ganules can bring TC and TG down and thus has a significant difference from that in the model group(p<0.01).When TC and TG are declined in each dose group of Simvastatin tablets and Qingzhi Tongmai granules,there appears a significant difference between groups(p<0.05); however,there is no obvious influence over the expression of LDL-C andHDL-C(p>0.05).2 Detection results of inflammatory factor in rabbit blood serum show that:compared with the blank control group, expression level of IL-6 and TNF-α in model group has an incredible increase(p<0.01).Each dose group of Simvastatin tablets and Qingzhi Tongmai ganules can bring the contents of IL-6 and TNF-α down and thus has a significant difference from that in the model group(p<0.01).There is a significant difference(p<0.01)when comparison is made between each dose group in Simvastatin tablets and Qingzhi Tongmai granules.Conclusion:1 After feeding rabbits with high-fat diet,contents of TC,TG and LDL-C has an incredible increase while HDL-C has an obvious decrease.It means that the rabbit model with hyperlipoidemia is created successfully.2 Contents of TC and TG in hyperlipoidemia group serum can be decreased apparently by Qingzhi Tongmai Granules, and the decrease tendency may become more obvious with the increase of the dose.However,Qingzhi Tongmai Granules is not as effective as Simvastatin tablets.Every dose group of Qingzhi Tongmai Granules doesn’t have obvious effect on the expression of LDL-C and HDL-C in serum.3 The expression level of IL-6 and TNF-αin hyperlipoidemia group serum is inhibited by Qingzhi Tongmai Granules,and the inhibition tendency may become more obvious with the increase of the dose.However,Qingzhi Tongmai Granules are not as effective as Simvastatin tablets.4 Qingzhi Tongmai Granules can prevent the functional mechanism of the rabbit’s atherosclerosis.Vascular endothelial cells may be protected by decreasing contents of TC and TG,adjusting blood lipid metabolism, inhibiting expression level of IL-6 and TNF-α and reducing AS inflammatory reaction.
