| Purpose:To observe the effect of Qingzhi Tongmai Granule on experimental hyperlipidemia rat model of lipid metabolism and the expression of the in-depth study of the best ratio of Qingzhi Tongmai granule in prevention and treatment of atherosclerosis,and observed between expression and blood lipid index of NPCI and NPC1L1 in liver and small intestine in contact.Material and method:1.the experimental animals selected were 108 male SPF SD rats.96 rats were randomly divided into 12 groups(normal group,model group and Chinese medicine group 10),with 9 rats in each group.After one week of adaptive feeding,the rats were modeled except the normal group.The model was injected with vitamin D3 + compound high-fat diet,and the normal group rats were fed with normal saline + common feed.2.After the end of the experiment at the 9th week,determination of total cholesterol(TC),triglyceride(TG),low density lipoprotein-cholesterol(LDL-C)and high density lipoprotein-cholesterol(HDL-C)content;the expression level of mRNA and protein of NPC1 and NPC1L1 in the small intestine and liver of rats was detected by PCR and Western blot.Results: 1.The results of blood lipid four items showed that: compared with the blank group,the content of TC,TG and LDL-C in the model group increased significantly(P<0.01).Compared with the model group,Chinese medicine group all decreased TG(P<0.01),3/8/9/12 group decreased TC content(P<0.05),the 8/10/11/12 group increased the content of HDL-C(P<0.05),decreased the content of 3/8/9/10/11/12LDL-C(P<0.05);group TC,TG and the rest of the content of LDL-C was decreased and HDL-C increased,but there was no statistical significance(P>0.05).2.Western-blot results showed that compared with the normal group,the protein expression of NPC1 in liver and small intestine decreased in model group and Chinese medicine group(P < 0.05),and the expression of NPC1L1 protein increased in liver and small intestine(P < 0.05).Compared with the model group,the expression of NPC1 in liver and small intestine increased in the Chinese medicine group(P < 0.05),and the protein expression of NPC1L1 in liver and small intestine decreased(P < 0.05).3.RT-PCR results showed that the mRNA expression of NPC1 in the liver and small intestine was decreased in the model group and the traditional Chinese medicine group compared with the normal group(P<0.05),and the mRNA expression of NPC1L1 was increased in the liver and small intestine(P< 0.05).Compared with the model group,mRNA expression of NPC1 in the liver and small intestine was increased(P<0.05),and protein expression of NPC1L1 was decreased in liver and small intestine(P<0.05).Conclusion: 1.it was confirmed that the lipid lowering effect of the eighth groups and the twelfth groups was better than the other traditional Chinese medicine groups.2.Qingzhi Tongmai Granules can increase the mRNA and protein expression of NPC1 in liver and small intestine tissue of model rats;NPC1L1 mRNA and protein expression is decreased.3.Qing Zhi Tong Mai Granule can reverse its cholesterol transport by increasing the expression of NPC1 and mRNA and protein in liver and small intestine,and decreasing the expression of mRNA and protein in NPC1L1.4.The mechanism of prevention and treatment of atherosclerosis through Qingzhi Mai Granule,probably by mRNA and protein expression of NPC1 increased in liver and intestine,reduce the expression of mRNA and NPC1L1 protein,and inhibit the absorption of cholesterol,delay the formation and development of atherosclerosis. |
