Purpose:To observe the effects of different compatibility of ingredients of Astragalus and Pueraria on liver glucose and lipid metabolism in diabetic rats, aiming to analyze the main effects and interactions of Astragalus flavonoid, Astragalus saponin and Pueraria flavonoid on this process in diabetic rats.Material and method:The 146 SD rats were randomly divided into 10 groups based on blood glucose, which namely(1) normal group,(2) model group,(3) positive control(Jinqi metformin) group,(4) Astragalus flavonoid group,(5) Astragalus saponin group,(6) Pueraria flavonoid group,(7) Pueraria flavonoid & Astragalus saponin group,(8) Astragalus flavonoid & Astragalus saponin group,(9) Astragalus flavonoid & Pueraria flavonoid group,(10) Astragalus flavonoid & Pueraria flavonoid & Astragalus saponin group, 15 rats in each group,except normol group(11 rats). In addition to the normal group, the other rats were given to make after the DM model by STZ(46 m g·kg-1) ip. Except the normal group and the positive control group, rest of the groups were implemented experiment according to 2×2×2 factorial design. After administration 30 days, contents of fasting blood glucose, serum cholesterol(CHO) and triglyceride(TG) were tested; the levels of hepatic insulin(INS), adiponectin(APN), leptin(LEP), glucose transporter 4(GLUT4), interleukin-12(IL-12) and interleukin-15(IL-15) were tested by enzyme-linked immunosorbent assay(ELISA); the expression levels of adiponectin receptor 2(Adipo R2) and peroxisome proliferator-activated receptor α(PPARα) m RNA in liver were detected by Reverse transcriptase-polymerase chain reaction(RT-PCR). Experimental datas were analysed through statistical software SPSS17.0.Results: 1. The weight of rats in the model group was significantly lower than normal rats, a slight increase was showed in weight of rats in treated groups. 2. The level of blood glucose were significantly increased in model rats, while compared with the model group, that was lower significantly in Astragalus flavonoid group, Astragalusflavonoid & Pueraria flavonoid group, Astragalus flavonoid & Pueraria flavonoid & Astragalus saponin group, and the difference was statistically significant(P<0.05). 3. Contents of serum CHO and TG were significantly increased in model group.(1) On level of serum CHO: Astragalus flavonoid, Astragalus saponin and Pueraria flavonoid could effectively reduce the levels of serum CHO and TG; a interaction shew between Astragalus flavonoid and Astragalus saponin, that was the combination worse than Astragalus saponin only; interaction effect also shew between Astragalus flavonoid and Pueraria flavonoid, the combined effect was worse than any single drug, the same with Astragalus saponin and Pueraria flavonoid.(2) On content of serum TG: Astragalus flavonoid could effectively reduce level of serum TG. And interaction shew between Astragalus flavonoid and Astragalus saponin, combination worse than Astragalus flavonoid only. Interaction also shew between Astragalus flavonoid and Pueraria flavonoid,the combined effect was worse than any single drug, and the same with Astragalus saponin and Pueraria flavonoid. 4. In model rat, levels of hepatic INS, APN, LEP and GLUT4 were significantly reduced.(1) On the level of the hepatic INS: Astragalus flavonoid, Astragalus saponin and Pueraria flavonoid could not enhance the level of hepatic INS; and there was interaction in these two drugs,the combined effect was better than any single drug;(2) On the level of the hepatic APN: the effects of Astragalus flavonoid, Astragalus saponin and Pueraria flavonoid were not obvious, there was no interaction between the two;(3) On the impact of the hepatic LEP level: Astragalus flavonoid could effectively improve the level of LEP; interaction of Astragalus flavonoid and Pueraria flavonoid better than single drug;(4) On the level of hepatic GLUT4: Pueraria flavonoid could effectively improved the level of GLUT4; Astragalus flavonoid and Astragalus saponin had interaction, combination was worse than Astragalus flavonoid alone; Astragalus saponin and Pueraria flavonoid had interaction, Pueraria flavonoid alone was better than combination; Astragalus flavonoid and Pueraria flavonoid had interaction, combination was better than any single drug.The secondary interaction among Astragalus flavonoid, Astragalus saponin and Pueraria flavonoid statistically significant, the combination of three drugs was worse than Astragalus flavonoid and Pueraria flavonoid compatibility. 5. In model group rat, the level of liver IL-12, IL-15 significantly increased.(1) On the IL-12 level in liver: Astragalus flavonoid could effectively reduced the levels of IL-12; there was no interaction between any two of them;(2) On the level of liver IL-15: Astragalus flavonoid, Astragalus saponin and Pueraria flavonoid could reduce the level of IL-15. Interaction was seen between Astragalus flavonoid and Pueraria flavonoid, combination better than any single drug; the secondary interaction of Astragalus flavonoid, Astragalus saponin and Puerariaflavonoid was statistically significant, the three drugs used in combination was worse than Astragalus flavonoid and Pueraria flavonoid compatibility. 6. Compared with normal group, the expression levels of liver Adipo R2, PPARα m RNA were significantly lower in model group rats, the difference was statistically significant(P<0.05). Compared with the model group, in Astragalus flavonoid & Pueraria flavonoid group, expression level of Adipo R2 m RNA was significantly increased; expression level of PPARα m RNA in Astragalus flavonoid group and Astragalus flavonoid & Pueraria flavonoid group was significantly higher, the difference was statistically significant(P<0.05).Conclusion:1. Astragalus flavonoid could reduce the level of blood glucose in diabetic rats significantly; Astragalus flavonoid, Astragalus saponin and Pueraria flavonoid could lower the levels of serum CHO, TG in diabetic rats, but had not shown the synergies. 2.Astragalus flavonoid and Pueraria flavonoid was the best combination on regulating glucose and lipid metabolism in liver of rats,because of their synergies in coregulating a number of indicators, such as liver INS, LEP, GLUT4 and IL-15. 3.Compatibility of Astragalus flavonoid, Astragalus saponin and Pueraria flavonoid could lower levels of blood glucose and lipid in diabetic rats, which may be related to the signal transduction of APN/Adipo R2/PPARα.
