Purpose: To observe the changes of plasma NO and ET the beagles dogs with myocardial ischemia were treated with Shengmai/Shenmai injection. Study difference the effects of Shengmai/Shenmaion endothelial function in myocardial ischemia model of beagle dogs and the optimal dose Shenmai Injection.Material and method: 42 healthy Beagle dogs with weight 8-10 kg were divided into the blank group, model group, low Shengmai injection group(0.8ml/kg), middle Shengmai injection group(1.6ml/kg), high Shengmai injection group(3.2ml/kg), Shenmai injection group(2.72/kg), western medicine by random. Every group had 6 dogs. Male and female is,Blank group dogs without treatment, normal food, water. Other groups in accordance with the 1.1 mg/kg dose given to beagles subcutaneous injection of isoproterenol, building mode for medicine 3 times, respectively, on the afternoon of the first day was administered once, second days morning third doses, caused by myocardial ischemia canine model. The last model administration completed at the same time, each group beagle dogs began to liquid uniform intravenous drip(corresponding dose of Shengmai injection, Shenmai injection, western medicine positive control drug: propranolol(5%) Glucose Injection 100 ml dilution), infusion time 1.5h.The Beagle dogs were observed before and after treatment of heart rate, ECG ST segment depression situation. Enzyme linked immunosorbent assay for the detection of myocardial ischemia after 1h, 4h, 8h, ET in serum, the content of NO, and compared.Results:1.Shengmai / Shenmai Injection on Beagle canine myocardial ischemia model ofheart rate, ECG. Myocardial ischemia model was successfully beagle dogs with elevated blood pressure and heart rate,or ECG ST segment depression. After the treatment of Shengmai Injection(high,middle,low) group of beagle dogs drop in heart rate, improved with different degree of ST segment in ECG, compared with the model group have significant difference(P<0.05) After the treatment of Shenmai injection group of beagle dogs rate drop, there are different degrees of improvement of ECG ST segment, compared with the model group have significant difference(P<0.05); compared with no difference in Shengmai injection group(P>0.05). After treatment, the control group of Western medicine in beagle dogs decreased heart rate, ECG ST segment is improved in different degree, compared with the model group were significantly different(P<0.05); and Shengmai / Shenmai injection group had no significant differences compared(P<0.05). 2、Shengmai / Shenmai Injection on Beagle canine myocardial ischemia model of NO content in serum. Myocardial ischemia model was successfully content of NO in serum decreased in Beagle Dogs。 The Shengmai Injection(high,middle,low,) content trend group Beagle canine serum NO the same. After treatment of 1h began to increase after treatment, 4h reached the peak, and then began to slow down, but the content of NO is still higher than that in the model group, the state of sustainable to elevated 8h. Shengmai injection group comparison, the content of NO in middle dose group, high dose group, low dose group. Inter group comparison of Shengmai injection of high, middle dose group than in the no statistical significance(P>0.05); With the low dose group than there are significant differences(P<0.05), Compared with the model group, Shengmai Injection(high,middle, low) group of each time point were higher than those in the model group, there was significant difference between the two,(P<0.05).By Shenmai injection treatment, the content of NO in serum in beagle dogs after treatment began to increase after treatment 1h, 4h reached the peak, and then began to slow down, but the content of NO is still higher than that in the model group, the state of sustainable to elevated 8h. Compared with the model group, Shenmai injection in each time point were higher than those in the model group, there was significant difference between them(P<0.05). And Shengmai Injection in high, middle dose group than no statistical significance,(P<0.05). And low dose group than there are significant differences(P<0.05) The western medicine propranolol treatment, the content of NO in serum in beagle dogs after treatment, 1h began to increase, and reached the peak at 4h, and then start to decline. In model group, and each time point than, no significant difference(P>0.05);decreased expression compared with Shengmai / Shenmai injection of NO at each time point, there are significant differences(P<0.05). 3.Shengmai/ Shenmai Injection on Beagle canine myocardial ischemia model of ET content in serum. Myocardial ischemia model was successfully content of ET in serum raised in Beagle Dogs。 The Shengmai Injection(high,middle, low,) content trend group Beagle canine serum ET the same. After treatment of 1h began to decreased after treatment, 4h reached the trough then began to slow raised, but the content of ET is still little than that in the model group, the state of sustainable to elevated 8h. Shengmai injection group comparison, the content of ET in middle dose group, high dose group,low dose group, inter group comparison of Shengmai injection of high, middle dose group than in the no statistical significance(P>0.05); With the low dose group than there are significant differences(P<0.05), Compared with the model group, Shengmai Injection(high,middle, low) group of each time point were little than those in the model group, there was significant difference between the two,(P<0.05). By Shenmai injection treatment, the content of ET in serum in beagle dogsafter treatment began to decreased after treatment 1h, 4h reached the trough, and then began to slow rise, but the content of ET is still little than that in the model group, the state of sustainable to elevated 8h. Compared with the model group, Shenmai injection in each time point were little than those in the model group, there was significant difference between them(P<0.05). And Shengmai Injection in high, middle dose group than no statistical significance(P>0.05), and low dose group than there are significant differences(P<0.05) The western medicine propranolol treatment, the content of NO in serum in beagle dogs after treatment, 1h began to decreased, and reached the trough at 4h, and then start to raised. In model group, and each time point than, no significant difference(P>0.05); higher expression and Shengmai / Shenmai injection of ET at each time point, there are significant differences(P<0.05).Conclusion:1. Shengmai / Shenmai injection can raise the level of NO, lower the level of ET. It indicates that Shengmai / Shenmai injection can treat CHD by improving vascular endothelial function. 2. Shengmai injection and Shenmai injection were not different on improving vascular endothelial function. 3.The middle Shengmai injection had effect on improving vascular endothelial function.
