Objective:In this paper, use Qi deficiency constitution mice for the study, aim to explore the curative effect and mechanism of HSY on anti-fatigue, anti-oxidation and the improvement of immune function.Methods:We used clinical manifestations of Qi deficiency constitution as standards, by comparing the modeling methods among exhaustive swimming method, standing in water method, free swimming+standing in water+depriving of some sleep method, took mice behavior, body weight, blood glucose (GLU), total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), swimming time ratio, liver glycogen content, lactate dehydrogenase (LDH) activity as indicators, screened out the animal model of Qi deficiency constitution which was in accordance with experimental requirements. The Qi deficiency constitution groups of low, medium and high dose were treated with HSY once a day at12.5g/kg,25g/kg and50g/kg, while the other groups were treated with physiological saline solution(equal normal saline).12consecutive days later, we observed the changes of mice in behavior and detected the weight, GLU, TC, TG, HDL-C, LDL-C, exhaustive swimming time ratio, liver glycogen content, lactate dehydrogenase (LDH) activity, superoxide dismutase (SOD) activity, glutathione peroxidase (GSH-Px) activity of spleen index, thymus index. Data were analyzed by SPSS17.0statistical software.Results:Qi deficiency constitution mice had less activity and showed fatigued compared with normal ones. Their weight, exhaustive swimming time ratio, liver glycogen content, LDH activity was significantly lower (P<0.01) while GLU was significantly higher (P<0.05) than normal ones. Treat with HSY after12days, the body weight, exhaustive swimming time ratio, liver glycogen, LDH activity, TG, TC, HDL-C, LDL-C of low dose group had no difference with normal control group (P>0.05), while the Glu, SOD activity, GSH-Px activity, thymus index and spleen index were significantly higher than normal control group (P<0.01), and the MDA significantly lower than the normal control group (P<0.01).The body weight, GLU、TG、TC、 HDL-C、LDL-C of medium and high dose groups had no difference with normal control group (P>0.05), while the exhaustive swimming time ratio, liver glycogen, LDH activity SOD activity, GSH-Px activity, thymus index and spleen index were significantly higher than normal control group (P<0.01). and the MDA significantly lower than the normal control group (P<0.01).With the dysfunction of lung, spleen and stomach, Zong Qi, Ying Qi and Wei Qi were decreased and caused body fatigue and immunity decrease which is the basis of Qi deficiency constitution of traditional Chinese medicine. The disorder of energy metabolism, such as sugar and fat, is the biological basis of Qi deficiency constitution. HSY can significantly reduce Qi deficiency constitution mice blood glucose and serum MDA content, improve the ratio of exhaustive swimming time, LDH activity, SOD activity, GSH-Px activity, spleen index, thymus index and improve Qi deficiency constitution mice function of anti-fatigue, anti-oxidation and immune regulation.
