Preparation Of Effective Part Of Psoralen And White Kernel And Study On Anti - Tumor Synergism

Study shows that psorslea corylifolia has anticancer activity, and compating with Fructus Ammomi Rotundus naphtha can further potentiate the anti-tumor effects of psoralen. The recipe has well-proven obvious inhibitory effect on the digestive tract tumors. Previous experiment shows that psoralen could strengthen the effect of anti-tumor ingredients in psorslea corylifolia, and eucalyptol is reported to be the main effective part of Fructus Ammomi Rotundus naphtha. This paper firstly studies the extraction and purification process of psoralen in psorslea corylifolia and the preparation of eucalyptol as the effective parts in Fructus Ammomi Rotundus naphtha. Next, research was performed to explore the antitumor synergy mechanism, from the perspective of pharmacokinetic interaction. The major results are as follows:1. Leaving to infuse for four times with 50% ethanol, recycled ethanol, heat with petroleum ether extraction of the lower black for three times, using petroleum ether extraction of the upper mixed suspension three times, can transfer much psoralen in psorslea corylifolia slices, and conducive to scurfpea fruit monomer purification, improve the extraction and purification efficiency of psoralen.The selection process of CO2 supercritical fluid extract psoralen as follows:1.5 times volume of 95% ethanol as entrainer slices,65℃,20M Pa, extraction of 2 h.2. The best extraction process of the effective parts of Fructus Ammomi Rotundus naphtha is medicinal materials powder for 24 mesh, do not soak, and nine times volume of water, extracting five hours.3. In vitro pharmacodynamic experiments show that psoralen has different degree of inhibition to the growth of human gastric cancer cells of MGC-803,BGC-823,SGC-823,MKN-45, and the inhibition has a time-dose-response relationship.The gastric cancer cell line SGC-7901 has the best inhibitory effect when psoralen and eucalyptol in 72:36μg/mL.4. LC-MS/MS method was established for the quantitative determination of psoralen in biosamples from rat. The method wasvalidated concerning the specificity, accuracy and sensitivity of this method. Based on this method, the pharmacokinetic results demonstrated thateucalyptol pharmacokinetic study can to a certain extent, to promote the absorption of psoralen in normal rats.5. Tissue distribution study shows that psoralen has distributed into well-perfusioned organs of the rats, predominantly in the liver, lungs, stomach, Compating with eucalyptol can greatly increase the psoralen in heart, lung, stomach, intestinal organs.
