Tumor is a disease which is seriously threatening human health and life, theincidence and mortality are increasing over the years, it’s lethality ranks second onlyto cardiovascular and cerebrovascular disease, the incidence populations has a trendtowards to youngers as well. At present, the treatment of malignant tumors are mainlybased on surgery, radiotherapy, and chemotherapy, although these methods havesignificant anti-tumor and curative effect, but they also bring serious toxic side effectsto patients. So looking for a low toxicity, safe and effective anti-tumor drug oradjuvant anti-tumor drug is important significance.Research shows that the tumor development is closely related with the freeradicals. MDA is produced in the process of lipid peroxidation, it is expressed highlyin ovarian tumors, lung cancer and breast cancer, it can cause protein denaturation andgene mutation, which promote the development of tumor.SOD can reducing the MDAdamage to the body through blocking lipid peroxidation reaction and reduce thecontent of MDA.Low Immune function is also an important factor in tumor formation. Thymusand spleen are body’s important central and peripheral immune organs, these organscan reveal whether the immune system functions condition. IL-2is a cell proliferationfactor which is produced by activated helper T cells, it can promote the proliferationof T lymphocytes and NK cells, and enhance their activity of killing cells, Mean whileIL-2can promote B cell to differentiate, proliferate and produce antibodies. TNF-α, abioactive factor produced by monocytes and macrophages, can directly kill tumorcells and induce the apoptosis of tumor cells.Phellinus igniarius of Birch is a precious medicinal fungus. Recent studies foundthat it has not only treatment menoxenia, overtiredness, hematuria, metrorrhagia,nosebleed, but the functions of reducing the content of blood sugar and protecting theliver, also it has antioxidant, anti-inflammatory, enhancing immunity and anti-tumor effect. People have been interesting in Phellinus igniarius of Birch anti-tumor effect.Objective: To investigate the anti-tumor effect of EBPI and explore thecorrelation between its anti-tumor mechanisms and SOD, MDA, IL-2, TNF-α andimmune function by culturing the cell line of H22、S180、HepG2invitro and implantH22、MFC、S180inmouse in vivo.To evaluate the possible anti-tumor effect andprovide a reasonable、safe、 effective theoretical and methodological basis forexploiting the new anti-tumor or adjuvant antineoplastic drugs.Methods: H22-bearing, MFC-bearing, S180-bearing, and S180ascites tumors micemodels were established, and with the use of the first three models, the research wasdivided into six groups: normal control group, model group, CTX group, high-(2g/kg), middle-(1g/kg) and low-dose (0.5g/kg) of EBPI groups, for S180ascites tumorsmice model was divided into five groups, without normal control group. Then the lifespan prolongation of ascites tumor mice、tumor weight、weight of mice、and immuneorgan indexes were determined. The we investigated EBPI’s effect on the content ofserum SOD, MDA of transplanted tumors mice.With the method of ELISA, weinvestigated EBPI’s effect on interleukin-2(IL-2) and tumor necrosis factor(TNF-α)levels of S180-bearing mice. Effects of EBPI on the Lymphocyte transformation andgrowth of H22、S180、HepG2cell lines were evaluated by MTT; The number ofperipheral blood granulocytes and lymphocytes were detected by blood smears andlevel of white blood cell by cytometry.Results:1.Inhibitory effect of EBPI on growth of H22、MFC、S180cell lineswere cultured in vivo: after injecting H22、 MFC、 S180tumor cells in thesubcutaneously, the EBPI groups in eating and mental state behave better than themodel group, the average tumor weight of EBPI groups was lower than that of modelgroup, especially medium and high dose group have obvious results(P<0.05,P<0.01). Inhibitory rate on H22was38.27%、47.96%、55.61%; Inhibitory rate onMFC was44.83%、47.7%、51.15%. Inhibitory rate on S180was38%、48.4%、53.6%;After administration of5d of mice can touch the tumor in the body. and on7d,8d,9d,10d for groups of EBPI, and CTX group the volume of tumor was smaller thanthat of the model group; meanwhile EBPI can prolong the survival time (P<0.05) andimproved living quality of S180ascites tumor mice. EBPI had no inhibitory effect on H22, S180tumor cell in vitro(P>0.05), EBPI only had inhibitory effect on HepG2cells at concentration of20~40μg/mL.2. The effects of EBPI on SOD and MDA of H22, MFC and S180-bearing mice:Compared with the model group after treatment with EBPI, the content of serum SODof H22, MFC, S180-bearing mice was significantly increased (P<0.05,P<0.01),thecontent of MDA was decreased(P<0.05).3. Effects of EBPI on immune function: Compared with model and CTX groupsthe thymus and spleen indexs of the EBPI group were increased significantly(P<0.01), The liver index was not increased significantly(P>0.05). EBPI canincreas the serum levels of the IL-2and TNF-α of S180-bearing mice(P<0.05,P<0.01). Obviously EBPI promoted the proliferation of mice’s spleen lymphocytes.Compared with CTX group, EBPI could increase granulocyte and lymphocyte countsin the peripheral blood of S180-bearing mice, without affect on the counts of normalwhite blood cells.Conclusion:1. In vitro, EBPI has no obvious inhibitory effects on H22and S180tumor cell, In vivo, it can significantly inhibit the growth of H22hepatoma cells,MFC gastric cancer cells and S180sarcoma cells, It can also improve survival state andprolong life span of S180ascites tumors mice.2. EBPI can increase thymus and spleen indexes of H22、MFC、S180-bearingmice,and induced IL-2and TNF-α generation in vivo and stimulate the proliferationof spleen lymphocytes of S180-bearing mice.3. EBPI can increase the counts of the peripheral blood granulocyte andlymphocyte, and play a role against mice peripheral blood leukocyte reductioninduced by CTX.4. EBPI can increase the serum activity of SOD and reduce the serum level ofMDA of solid tumor mice its anti-tumor mechanism may relate antioxidant ability andimproving body immunity.
