A Comparative Study On The Relationship Between The Processing Of Ershen Pills And Their Spectral Effects

Processing of traditional Chinese medicine and compound medicine are the two major characteristics of Chinese medicine.Semen psoraleae and nutmeg before and after processing have been respectively combined into compound Ershen Pill and then four kinds of Ershen Pill combination have been formed.By HPLC fingerprint and GC-MS technologies,the influence of processing on the chemical compositions of Ershen Pill have been fully analyzed.And the effects of warming the spleen to stop diarrhea have been compared by efficacy experiments,in order to select the best Ershen Pill combination. Using the best Ershen Pill combination as a proxy.five parts has been separated by the means of systematic solvent extraction. Afterwards, the HPLC fingerprints of different parts have been established and the effects of warming the spleen to stop diarrhea have been compared.By correlation analysis, the spectrum effect relationship has been established to locate the chromatographic peaks which contribute to the effect.Finally the method of LC-MS has been used to determine these effective components.The experimental results show that the retention times and peak areas of fingerprints and GC-MS total ion current chromatographic peaks of the four kinds of Ershen Pill combination are partly different.That means there can be differences in the kinds and contents of the chemical compositions. And these differences can lead to the efficacy divergence.Ershen Pill â…£ which is composed of psoralea stir-fried with salt and nutmeg baked with bran.can significantly reduce the diarrhea index of acute diarrhea mice caused by senna as well as intestinal propulsion rate and gastric emptying rate of healthy mice.It indicates that the drugs in Ershen Pill should be processed to incruce the effect of warming the spleen to stop diarrhea.Namely psoralea should be stir-fired with salt and nutmeg baked with bran when Ershen Pill is used to cure dawn diarrhea caused by deficiency of spleen-yang and kidney-yang.The effect of Ershen Pill â…£ originates in the low polarity components,and the effect of the petroleum ether is the most obvious.By establishing the spectrum effect relationship, the chromatographic peaks which contribute to the effect can be located,including No.6>18>7>20>14>21>1>25>24>23>4>3>8>11.And the correlation of these peaks are close. As the volatile components of Ershen Pill are closely related to its effect,GC technology still need to be applied to the research of the spectrum effect relationship,in order to locate the volatile components which contribute much to the effect.By LC-MS technology,these peaks can be preferred.peak No.3 as psoralen, peak No.4 as isopsoralen.peak No.7 as neobavaisoflavone.peak No.8 as corylifolin,peak No.11 as myrisisolignan,peak No.14 as corylifol D,peak No.18 as isobavachalcone,peak No.20 as bavachinin,peak No.21 as corylifol A.peak No.25 as bakuchiol.In addition,peak No.1 and No.6 may be composed of two or more kinds of components.And peak No.23 and No.24 are still unknown.The determination of effective components still wait for further research by nuclear magnetic resonance and other technologies.The effect of warming the spleen to stop diarrhea of Ershen Pill â…£ may come from the common role of a variety of components.
