This paper can be divided into three chapters:The first chapter describes the HPLC method for simultaneous determination of methyleugenol,safrole and asarinin in Asari Radix et Rhizoma.The second chapter describes the influence of processing on the contents of safrole, methyleugenol and asarinin in Asari Radix et Rhizoma.The last chapter describes the variation patterns of safrole, methyleugenol and asarinin in Asari Radix et Rhizoma at different decocting time.Herba Asari is a traditional Chinese medicinal herb commonly used as a crude drug and an ingredient in patent medicines.The main active ingredients of the herba contain volatile components and non-volatile components.The volatile components contain methyleugenol,safrole,elemicin, a-pinene,(3-pinene and so on. And the non-volatile components have many compounds,such as asarinin,sesamin and kakuoLAmong the components as mentioned above, methyleugeno and safrole is not only the active ingredient,but also the toxic components. The asarinin has antivirus and anti-mycobacterium tuberculosis effects.In2010edition of Chinese Pharmacopoeia the determination of asarinin is used as a quality control standard for the herba Asari.So far there are many studies on the herba Asari,such as the determination of methyleugenol and safrole by HPLC or GC, and the determination of asarinin by HPLC etc.But there are not studies on the simultaneous determination of both the volatile components and the non-volatile components.Conclusively,this research develops an HPLC method for simultaneous determination of methyleugenol,safrole and asarinin in Asari Radix et Rhizoma,which can be used for the quality control standard in the herba Asari.The Compendium of Materia Medica records that the dosage of herba Asari must be small because of its slightly toxicity.By retrieving literature,we comprehend that there arc many processing methods of herba Asari,such as "bake to dark brown"、"baking dryness",which can reduce toxin and relieve drug nature.Consequently,this research studies the influence of processing on the contents of safrole,methyleugenol and asarinin in Asari Radix of Rhizoma,and researches its rationality of processing,which can provide the reference method for the processing of herba Asari. One of the Traditional Chinese Formuloations,the herbal decoction is the most widely used in the Traditional Chinese Medicine clinical.So most of practitioners and scholars hold that the methods of administration of herba Asari are always decoction.So this paper determines directly the contents of safrole,methyleugenol and asarinin in the decoction of herba Asari from30minutes to120minutes by HPLC,to reflect the content changes of main toxicity composition and effective component We hope that this research can provide the reference method for the safety of herba Asari on the clinical medication.
