Study On Online Expert System For Chromatographic Fingerprinting Of Traditional Chinese Medicine

The expert system of chromatographic fingerprint on line is combined the analytical chemistry,the science of chemometrics with the computer network technology,and so on.Also, the expert system can guide chemical workers design or select the optimal conditions.We used the establishment of the expert system for HPLC-fingerprint on line as an example.The fingerprint chromatograms were based on the system.1.We used the establishment of the expert system for HPLC-fingerprint on line as an example.Establishment of the working memory,by analysis,sorting the ralated information of the HPLC,which we extracted the necessary material,we transformed them into data according to a certain format.Then they were stored respectively in total of 6 tables as database.Currently it is filled with 21 HPLC fingerprints of traditional Chinese medicine,which could be appended later.Establishment of the knowlege base and the inference mechanism,through the literature analysis,reasoning and the process of summing up,it gradually formed a rule.Three ways which were general approach,families and major types of compounds were established.Families method was based on the idea that the same family of herbs had similar material,and established by analysis of each commonly used representative herbs’ chromatographic conditions in literature and summarizing their commons.Major types of compounds method was based on the idea that same type of compounds generally had properties in common,and established by analysis of each type of compounds’ chromatographic conditions in literature and summarizing their commons.General approach was used when users didn’t find the right results what they wanted in the other two methods,or users weren’t satisfied with the other two methods.This method was actually simulating experts obtaining a fingerprint chromatogram of an herb.2.The fingerprint chromatograms were based on the system.The scientific and reliability of this system was verified by its application in experiment.Establishment of HPLC-fingerprint analysis of Radix Glycyrrhizae:The chromatographic condition included Kromasil C₁₈column,the gradient elution with acetonitrile and 0.05% phosphate acid solution.The concentration of acetonitrile increases from 15%to 20%in 5 minutes,increases to 25%during the later 20 minutes,increases to 90%finally at the 80 minutes. The detection wavelength was 276nm.Establishment of HPLC-fingerprint analysis of Atractylodes macrocephala Koidz.:The chromatographic condition included Kromasil C₁₈column,the gradient elution with methanol and 0.05%phosphate acid solution.The concentration of methanol increases from 5%to 70%in 30 minutes,keeping 70%during the later 30 minutes,increases to 100%during the later 15 minutes,keeping 100%finally at the 90 minutes.The detection wavelength was 280nm.
