Objective Through a series of comparative studies with the endangered species of Atractylades Macrocephalacv.Yuzhu. and Atractylodes Macrocephala Koidz., including character comparison, microscopic identification, TLC and chemical components comparison, clarified the genuineness of Atractylades Macrocephalacv.Yuzhu., Establish the characteristic chromatogram and rapid identification of infrared spectrum to control the quality of Atractylades Macrocephalacv.Yuzhu., and provided basis for GAP, protection and development of it. Methods Microscopic identification, TLC identification and HPLC analysis were used to set up a preliminary identification and establishment the characteristic chromatogram; near-infrared spectroscopy was developed for establish the rapid detection model and experimental analysis of atractylenolide Ⅰ, atractylenolide Ⅱ, atractylenolide Ⅲ, atractylodin. Results Though the method of microscopic identification, stone cells, fiber and duct structure is similar to each other, but less inulin and more needle-like crystal, stone cell in Atractylodes Macrocephala Koidz.; they both appeared pink main sopt at the same time and same positon by using TLC; HPLC analysis showed that the content of atractylenolide Ⅰ was high and atractylenolide Ⅱ, atractylodin were low in Atractylades Macrocephalacv. Yuzhu., the content of atractylenolide I, atractylenolide Ⅱ, atractylenolide Ⅲ, atractylodin were high in Atractylodes Macrocephala Koidz. from Xinchang,the content of atractylenolide Ⅰ and atractylenolide Ⅲ were high in Atractylodes Macrocephala Koidz. from Panan, the content of atractylenolide Ⅰ, atractylenolide Ⅱ, atractylenolide Ⅲ were low in Atractylodes Macrocephala Koidz. from Suntown. Through the study on the characteristic chromatogram, there are 21 common peaks, and identify the 6,12,14,16 peaks were atractylenolide Ⅲ, atractylenolide Ⅰ, atractylenolide Ⅱ, atractylodin. The results of the rapid identification modle of Atractylades Macrocephalacv,Yuzhu. and Atractylodes Macrocephala Koidz. by near-infrared spectroscopy analysis were accurate, showed the model was reliable. Establish the quantitative calibration model of the total content of atractylenolide Ⅰ, atractylenolide Ⅱ, atractylenolide Ⅲ and atractylodin in Atractylades Macrocephalacv.Yuzhu. and Atractylodes Macrocephala Koidz. by using partial least squares, the model can be used for the rapid detection of total content of atractylenolide Ⅰ, atractylenolide Ⅱ, atractylenolide Ⅲ and atractylodin in Atractylades Macrocephalacv.Yuzhu. and Atractylodes Macrocephala Koidz..Conclusion This research clarified the genuineness of Atractylades Macrocephalacv. Yuzhu., provided scientific reference for the further study and resources development.
