Experiment was conducted with the young leaf and petiole in vitro of 'Nubiana' (Prunus salicina) for studying the effects of different basic culture media, hormone levels, agar content, organic addition on the callus inducement,subculture and plant regeneration. And the effects of hormone levels, culture time and agar content on the adventitious plant regeneration of 'Nubiana' were also studied. The main results were as follows:Firstly, the optimal explants for callus induction of 'Nubiana' were leaves and petioles.Secondly, the optimal medium for leaves and petioles callus induction of 'Nubiana' was WPM+2,4-D 2.0 mg·L-1+6-BA 1.0 mg·L-1+glucose 30.0 g·L-1+agar 5.0 g·L-1. Selecting explants which had been sub-cultured around 28 d, the best duration of dark was 28 d.Thirdly, the optimal medium for callus subculture of 'Nubiana' was LP+2,4-D 1.0 mg·L-1-BA 0.5 mg·L-1+sucrose 40.0 g·L-1+CH 0.4 g·L-1+agar 6.0 g·L-1. Selecting explants which had been sub-cultured around 21 d, the callus was sub-cultured in darkness all alone.Fourth, the optimal medium for callus regeneration of 'Nubiana' was WPM+NAA 1.0 mg·L-1+6-BA 0.5 mg·L-1+TDZ 2.0 mg·L-1+sucrose 30.0 g·L-1+agar 6.8 g·L-1, by 6.7% of adventitious bud rate. The callus was sub-cultured under white light.At last, the optimal medium for petioles culture of 'Nubiana' was WPM+TDZ 0.5 mg·L-1+sucrose 30.0 g·L-1+agar 6.0 g·L-1. Selecting explants which had been sub-cultured around 21 d, the best duration of dark was 14 d, by 35% of adventitious bud rate. Adventitious shoots were mainly regenerated at proximal cutting region.
